No one has ever seen Mr. Samberg and me in the same room at the same time. Probably a coincidence though...
The "want-need" pair is probably something that aspiring writers know intuitively, but unless they've taken some courses or studied writing theory, it's probably not something they think about explicitly. Thinking on my own writing, when I get stuck it's because I've not addressed the protagonists' wants and needs.
100% correct that it’s something I intuited but I hadn’t quite connected the dots in this way. A lot of emphasis is put on the want but not as much is put on the need.
The concept absolutely speaks to the human condition as we often want things or think we do but we don’t look our lives objectively enough to recognize that what we want is often not what we need and even contrary to what we need.

Do you have any other character examples built from this method?
Even though Iron Man didn’t die, the fact he was willing to sacrifice himself for the greater good is satisfactory, in my opinion. This demonstration of a change to a more heroic attitude completes his arc of earning redemption, developing humility, and absolving his guilt.
Sure, sequels are always inevitable, but even without business and from a pure writing standpoint, not dying at the end allows this character future opportunity to grow and develop in new ways.
The funny thing is that even with Tony Stark, his flaw stays the same. The issues manifest in different ways, but ego is deeply rooted in his character. In Iron Man 2 for example, there is the entire plot about Tony not wanting anyone else to have his tech. In the end though, he only saves the day with help from War Machine. In that same way, he needed to overcome his ego to take the day. Only in this case, it's a bit more flimsy, but Iron Man 2 isn't anywhere near as tight as Iron Man 1 narratively speaking.
As for another example that isn't Tony, there are plenty out there. In the Stormlight Archive, Kaladin needs to overcome his depression to get what he wants, to help people. In Star Wars, Luke needs to learn to trust the force in order to destroy the Death Star. Luke's flaw isn't as glaring as Tony Stark's, but they establish early in A New Hope that he thinks the Force is hogwash. That's still a flaw in the narrative sense.

Any tips for giving a character a strong "wan't" beyond "survive" if the meat of the story (or at least the beginning) involves running away from deadly pursuers?
My story in a nutshell is a fantasy story where the protagonist finds out she's the illegitimate daughter of an emperor overthrown before her birth and most politicians want her dead so she doesn't pose a threat to their political power.
Wants that go beyond fundamental stuff like survival can really come from anywhere! Like in your story, it seems like your main character wants to live a normal life. Or she can want to destroy those politicians. Those two different wants are going to give you WILDLY different character traits and propel the story in different ways.
