Amlodipine (Norvasc) belongs to class of calcium channel blockers, known as dihydro-pyridines. Amlodipine is used to treat hypertension or high blood pressure, chronic stable angina or chest pain, and coronary artery disease. Amlodipine has a very long half-life and does not require a sustained-release formulation.
Amlodipine (Norvasc) works by affecting the movement of calcium into the cells of the heart and blood vessels. This relaxes the blood vessels and lowers blood pressure, and increases the supply of blood and oxygen to the heart while reducing its workload.
Common side effects of amlodipine include: headache, dizziness, flushing of the skin, peripheral edema, hypotension, reflex tachycardia, and gingival hyperplasia. Amlodipine is considered the safest, if a calcium channel blocker must be used to lower blood pressure, in patients with heart failure with reduced ejection fraction.
Use amlodipine with caution in Hepatic and renal disease. Amlodipine is contraindicated in Pre-existing bradycardia, heart block, and heart failure. Amlodipine has drug interaction with grapefruit and digoxin. Amlodipine is a major substrates of CYP450 3A4 enzyme. Strong CYP3A4 inducers and inhibitors should be used with caution, and in some cases, avoided in combination with amlodipine. Grapefruit juice should be avoided.
