Heating should be done for 10min before adding acetocarmine and glycerine,and that too in 1Normal HCl(instead of water or can use water),so that the stages are fixed and pre-arrested like suppose we have to see metaphase so that stage will be fixed and we can observe the specific stages easily

First heat the slide,then add acetocarmine,remove extra stain by blotting paper,then discard the light stained region and put the dark stained region in slide,then add glycerine to prevent overstaining
