Ed and Izumi were using alchemy to change/weaken the composition of Greed's armor before attacking various parts of him, which is why they were able to penetrate his skin. I couldn't tell if you just cut it from the reaction or if you missed it.
1:43 Just a small clarification - Ed lost his leg when he tried to bring back his mom, not his arm. He gave up the arm to get Al's soul back.
2:34 I'm pretty sure Alphonse knows Ed and Izumi both saw something indescribable and that it's the reason they dont have to use a transmutation circle and can simply clap their hands to perform alchemy.

5:40 Ok I see, you missed the connection between seeing the Truth and not having to draw the circles. Ed and Izumi can use alchemy without a circle purely because they've seen the Truth. It makes sense that you keep thinking the Truth is something that needs more delving into at this point in the story, because you missed the significance of it that was already explained.
