- Benzodiazepines are commonly used to treat anxiety disorders and sleep disorders. They are thought to exert their effects in the brain by acting at receptors for the neurotransmitter gamma-aminobutyric acid, or GABA. In this video, I cover the the mechanism of action for benzodiazepines.

Welcome to 2 minute neuroscience, where I explain neuroscience topics in 2 minutes or less. In this installment I will discuss benzodiazepines.
Benzodiazepines are a class of drugs named for their chemical structure that are commonly used to treat anxiety disorders and sleep-related disorders. They include well-known drugs like valium, xanax, and klonopin. There are dozens of drugs in the benzodiazepine class, but the mechanism by which they all exert their effects is thought to be similar.
The sedating and anxiety-reducing effects of benzodiazepines are believed to be attributable to the drugs’ actions at receptors for the neurotransmitter gamma-aminobutyic acid, or GABA. In particular, benzodiazepines act at a subtype of GABA receptors called the GABAa receptor; GABAa receptors that also bind benzodiazepines are sometimes called benzodiazepine receptors.
When benzodiazepines bind, or attach, to the GABA receptor, they bind at a location separate from where GABA itself binds, and exert an influence over GABA binding. This type of action is called an allosteric effect, and in the case of benzodiazepines it results in increased action at the GABA receptor. There is not complete consensus on exactly how benzodiazepine binding affects activity at the GABA receptor but there is evidence to suggest that it increases the likelihood that GABA binding will activate the receptor and/or increases the effect that GABA has when it binds to the receptor.
That effect is to open an ion channel and allow the passage of negatively charged chloride ions into the neuron. This influx of negatively charged ions pushes the membrane potential further from zero, or hyperpolarizes it, and makes it less likely the neuron will fire an action potential. This type of neural inhibition is the basis for the effects of benzodiazepines, for by inhibiting the activity of neurons that make up networks involved with anxiety and arousal, the drugs are able to produce calming effects.

Note to public: You should not take benzos for longer than a week to two weeks because you can easily develop a dependency on them and it will be very difficult for most to get off. They are great for on the spot anxiety busters like before a medical procedure or a panic attack, but I would highly caution against further use. They are a nightmare to get off, for so many. There are people who stop taking them and are fine, but you don't want to risk finding out which one you are. Russian Roulette.
Why cant i just take them for the rest of my life ?
Klonopin is definitely easier to get off of than Ativan. I'm tapering off Klonopin right now after a month or so. I take it every three days this week. It's not bad at all. Ativan is another story
@@wtbwrites1806 It’s all relative, WTB. Some people have an easier time with one over another but the truth is that they all can be brutal. Clonazepam has literally ruined my life: memory loss, cognitive impairment, can’t focus, can’t converse normally, blank mind, can’t work. I have heard the same from others who have taken clonazepam. But I’ve heard the similar horror stories from people trying to get off all benzodiazepines and other psych meds. It is truly insane how difficult it is to get off these drugs, and the lasting brain damage they can cause. I’m sorry you’re struggling as well and wish you the best in getting off the benzo, and wish you a full recovery.
@@balkanjoker3111 it can cause tolerance withdrawal, permanent neurotransmitter damage, memory loss, and dementia. Tolerance withdrawal is when you go through all the symptoms of benzo withdrawal while still taking the benzo. All these are documented outcome to staying on a benzo the rest of your life. The drugs now have a black box warning against prescribing for older patients because of dementia, so many are rushing to get their patients off them, which is proving difficult. Many doctors do not know how to safely get their patients off these highly “addictive” and brain-chemical altering drugs… they know how to write a prescription but not how to properly taper them off which could take years. It’s harder the longer you’ve been on them. Doctors used to leave patients on the drugs “for the rest of their lives” but it’s simply irresponsible, negligent and ultimately cruel.

For anyone taking benzodiazepines DO NOT quit cold turkey. You need to taper off very slowly or you could face a withdrawal that has the potential to kill you or cause seizures. It happened to me. Benzo withdrawal is hell and you must taper.

I felt this way..... and then I ran out of my script a week before I could refill it... Day 6 cold turkey; I'm in agony and I never want to take them again
I've been 23 years on Xanax but still my doctor knows better than me what type of effects they have although he's never taken a single Xanax or benzo pill himself. Year after year he insisted they had no harmful effect on me, cause he knew best. I stopped a year ago and have been in hell ever since. "See? You better take Xanax, you obviously need it" is my docs response. The loss of body control and the constant fear of everything and anything is the worst. The tremors, muscle dystonia, muscle tension from hell, feeling of being electrocuted etc etc etc.
I tapered off clonazepam after taking it as prescribed for 23 years. I was cut off cold once n had a seizure, my eyesight never repaired itself. My taper went well only destroyed my teeth but nothing like that cold cut off..during those 3 months of being cut off cold I was shocked how the medical field treated me..Like trash, it was real scary stuff. Clueless doctors n nurses. I feel for you! Hang in there!
@@NoName-pu5ls I still have all the same physical symptoms. Heavy tremors, muscle tension, movement disorders, fear, hyperacusis, extreme anxiety, body zaps etc. It's been over a year since I quit Xanax. Will this EVER go away?!
@@Underhills from what I've read from others, 2 years they feel fully healed. Noting brain healing is a snail's pace but does heal.

I've read about protracted withdrawal (Ashton Report) with people suffering from all kinds of physical and mental issues for several years after benzo consumption. I have no evidence that I'm actually experiencing withdrawal from all those years of Xanax cause no doctor or neurologist can confirm if its actually benzo causing the tremors and extreme muscle tensions etc. But my perception is that the medical industry don't know much about this at all. I strongly suspect Xanax as a cause of my physical problems. It's just terrible, it's like someone decided to rewire my brain and I can literally feel electric currency running through my system. The brain seems hyperactive, like neurons firing on all cylinders and I'm scared.
@@Underhills I would be scared too!! Only very few benzo wise doctors acknowledge PAWS. Petrifying experience. I had an ER doctor deny benzo withdraw, so all these expensive test were performed to no avail of course. So confusing. Wow! I sure hope you heal faster...🐶💗🌹

Been on klonopin 20 years and it helped save my life. They are very effective and you can off them slowly if necessary. Nothing else helps.

I have considered doing the cranial nerves many times. I haven't done it mostly because there are already so many resources available for learning the cranial nerves. But maybe at some point in the future. Thanks!
Why does UA-cam have so many negative comments on any drug? If it helps, it helps long term or temporarily. Everyone is made aware of the side effects and dependency, so it is fine.
Please post positive comments on alternatives and how to lean off it.
@@AdamMitchellv yes totally. This is the one drug that helped me,, helps me, so let it be. To each his own, nobody comes around commenting on stuff when it works. People only b*itch when something doesn't work.
Let me explain. Those negative comments need to happen especially surround benzodiazepines, you say your aware of the side effects and dependency issues... You have no idea, see you've probably google side effects of benzodiazepines and saw a cute list with maybe ten symptoms meanwhile Dr. Heather Ashton has noted in her practice of well over a 150 systems a person can have both while on the drug and coming off all the way into the recovery phrase which on average lasts anywhere from 9months to 4 years. My problem with posts like these is that you are dismissing the harm of benzodiazepines and chalking it up to "Oh well they must just be negative comments", no they are trying to warn you. There's a reason another black box warning was published Sept 23rd/2020 and there's a reason many doctors are losing their jobs, pharmacies are getting in trouble for filling scripts longer then recommended which is 2-4 max, there's a reason rehab programs are closing down their benzodiazepine programs finally because the success doesn't justify the failure rates and lastly there's a reason more people are speaking out against these drugs. There's much more you don't know about benzodiazepines but let's just say you are already a victim to them if you use them regularly or "As needed" longer then a month which is already far too long. I get that you think they're helping you which is fine, but at least educate yourself before dismissing others we are trying to help you. The biggest misconceptions we see in our groups is
1) They were helping me
Yeah in the beginning sure, however once dependency kicks in which is around 7-9 days of usage, the GabaA and Glutamate receptors along with your CNS/PNS start to weaken, these drugs that use to help keep your anxiety in check becomes the aggressor, it's making your anxiety worse we which requires more of the drug until you are trapped in a vicious circle. This class of medication is far worse then you could ever imagine
2) You only get harmed if you abuse it or are a addict lol
In that FDA black box warning it's been concluded that benzos are short term only therapies, 72% that we know of used the drug responsible with only a 12% actually abusing the medication.
3) Well I use them as needed so I'm safe.
Nope, in fact you have a bigger change of developing a neurological phenomenon called kindling because you are stopping and starting the medication again which means that symptoms will get worse and worse
4)Ive been on for decades, I think I'm fine now.
Nope, the longer you are on the more damage you've done to your CNS/PNS, again there's a reason doctors are speaking out against the horrors of these drugs and if you've been on longer then 7 years, your chances of getting off decrease as permanent damage sets in.
5) I'll only go through withdrawal if I stop the drug lol
Nope sorry but that's not true. Benzodiazepine carry a very sinister issue called "Tolerance withdrawal". It's when you reach a point where the body, GabaA and Glutamate no longer want anything to do with the drug and you stop feeling the effects of the drug, you body then goes into withdrawal. It doesn't matter how much you increase the drug you won't get anything from it. You will go into withdrawal and all the damages your neurological system has gained while on the drug, all the anxiety that's been increased while on the drug... You will have to deal with that for life now. You will make the mistake of going to the doctor or rehab to get off the drug but as within the same FDA black box warning on Sept 23rd/2020 states, your doctor can't recognize the symptoms of harm from the drug and rehab programs are closing their benzo doors because the failure rate is so high. You will be given another medication that effects GabaA which is a no no because those receptors need to recover on their own or your doctor will put you on another benzodiazepines... Why do you think so many are trying to warn you? To scare you lol
Benzodiazepines hit the market in 1960 with valium hitting the market in 1963,since then we've had nothing but horrorifc stories of long term disability due to the drug and get this, we don't have a single study that suggests its safe, it's beneficial and that it helps people longer then 3 months in fact we do have reports and documentation suggesting the person will get worse.
At least educate yourself instead of just excepting that these pills are helping you which is cowardly, you should be looking for other ways to take care of your anxiety and sleep issues... Otherwise you end up like us who can no longer take a anti anxiety pill ever now because of benzodiazepines. Look up Heather Ashton's work, go to Facebook benzodiazepines withdrawal groups and just learn something because things will get worse for you simply because you dismiss what you feel is uncomfortable to hear... Trust me, we've been there and now we wish we listened. We took those pills as prescribed and now our lives are destroyed. You are probably having symptoms that the doctor can't figure out which is being caused by the shock to your CNS/PNS and down regulated GabaA and Up regulated Glutamate receptors.

You think these benzodiazepines are he long you lol, they are down regulating your GabaA receptors and up regulating your Glutamate receptors causing a imbalance ontop of depressing and oppressing your CNS/PNS until one day they stop working. Heather Ashton has identified 150 a person can experience, there's a reason multiple black boxes exist, for some reason people believe they're special. The drug not only makes anxiety worse which is documented heavily and it stops working after a while. It's the reason 2-4 weeks is the maximum duration which you can call any pharmacy and ask. Read my other comment it has way more information but you are in for a terrible time. Good luck

Benzos were great until I stupidly mixed alcohol with klonopin over a weeks time and I turned into a crazy psycosis and then the wothdrawl from klonopin was miserable. It took me over 3 weeks to start to almost feel back to somewhat normalish. I only took bemzos for a month and a half, but obviously mixing them with alcohol is a horrible idea.
Did ylu get terrible brain fog? I got 1 time iv ativan and then lifes beem hell
@@syntholshoulders1842 no i had a devastating crash which lasted several weeks in which I went into withdrawl for both benzos/ alcohol. I was living life, drinking socially, not working, living my best life, then I had severe anxiety/ panic attacks whenever I left my place. Was terrible until I went away after some weeks.

It will. In my experience much more smoother than anti psychotic.
Same here well I'm Rx Klonopin nowadays but my Dr started me out on Xanax followed by Ativan (the worst drug I've ever had to come off of as far as benzodiazepines go) at least with Klonopin I can miss a couple of days and not feel immediate withdrawal (as I did with Aitvan/ Xanax) Also Valium was the smoothest for me. One 5 mg tab and I'm good for 4 to 5 days benzo free.
Nick John yeah it’s good for a while... then when you get of it. It’s the hell that starts. Please people before taking them do some research it’s not worth the withdrawal pain.

Klonazepam has halflife of 24 hours so you may go without for 48. lorazepam(ativan) lasts merely 8h. xanax is the shortest - 4h half life. goes away quick and withdrawal is sudden and intense unlike longlasting benzos like clonazepam diazepam. diazeopam is used to quit benzos because its smoothest wd
Now describe how benzodiazepine tolerance works; why the drug completely loses therapeutic affect. Administration of the drug makes you sick, but cessation of the drug kills you(or tortures you till you wish for death).

Thanks! I've thought of expanding the channel, but honestly I just don't have the time. This is just a hobby of mine and I only have limited time to devote to it. But it's not just about the views for me and as long as people are finding the videos useful in helping them to learn about neuroscience, then I'm satisfied.
Neuroscientifically Challenged well hey if it's just a hobby then by all means, keep doing what you're doing. They're amazing for epilepsy. I've had my fiancé save me after I told her exactly all the things that can help. I had a very fast acting diazapam thing. (that's far as ill go.. Lol)
Well.. I was in a lot of pain from breaking my back.. I wanted a hot bath. Had a seizure. She managed to get me out, give that.. Get an ambulance.
Too bad these medications are demonized so quickly. If used correctly, they work well for anxiety and other things.
There is no such thing as using benzos "correctly" other than one does prior to a surgical procedure. The risk of iatrogenic injury due to prescribed use of benzodiazepines is great.
@@THXx1138 i mean.. I get your point but if i wouldnt have had the possibility to take em i wouldve killed myself long ago. Ofc it should always be short them solution and moderated. Mal practice isnt rare. One psychiatrist wanted to give me a dose of 2mg a day - which is absolutely insane. I would be a zombie. Thankfully i hesitated on it and so did my mom and she told me to just take half.
Agreed when not abused and used as prescribed they can be miraculous for some peoples quality of life.
@@THXx1138 Exactly this. People don’t understand. Taken as prescribed these drugs cause iatrogenic injury. I took clonazepam as prescribed by my doctor for 7 years, became very ill while on them, but the withdrawal off of them has damaged my brain beyond anything I can accurately describe here. I am in several benzo-harmed support groups and there are thousands upon thousands of us out there- probably millions internationally, but hardly anyone knows about it or they just refuse to acknowledge it. People think you are only harmed if you “abuse” but this a total fallacy.

I don't think the true amount of people affected is known but it must be millions of people as you say and sadly not everyone researches the things maybe due to trusting doctors who don't know the true horror or maybe think it's exaggerated.
If anyone does use them please use for a short time but if physically dependant don't stop suddenly.
The fastest safe taper is 10% dose cut every two weeks but more reasonably 5%.
I won't say they are evil, even now but they can be for some people, even low dose short periods.
I can also verify its not just abusers who go through this.
The scary thing is Gaba related withdrawal is not predictable and linear like opiate withdrawal for example it has a clear peak and end point.
This withdrawal is random peaks.
Could be OK for 2 hours/days (Windows) then an emotional wreck for weeks or months.
It really can change hour to hour.
Each time you think that's it another wave hits, it teases and torments. That's why many say it's h*ll (OCD can't say that word lol) Tormented by your own body.

Great video. Theanine and Pharma GABA are good nutritional supplements to use instead of benzodiazepines. These supplements work in a similar with GABA receptors and are non-addictive. GABA Soothe from Jarrow formulas is a good choice.

I feel anxious. Can't sleep without the sleeping pills or anti anxiety drugs. And to make things worse I think I’m getting addicted to them. How can I make myself feel better and sleep better?
Excersize hard for first periode. After that the sleep will come back.
CBD and melatonin until you get back to normal.
@@thearchiveable that's a very vague answer to a very complex situation. I've had friends recommend the old "work out sleep better" stuff but what they maybe even you don't understand is once a chemical dependence develops no amount of physical activity will lessen the withdrawal process. Maybe in the longer run and on a life style basis but simply stopping these drugs cold and trying to go it alone is almost surely to fail anyone. I know, I've tried for nearly a decade now. I just wish I had know these long term effects before I ever started taking these medications.

Can "brain lock be a symptom from benzo and tapering? Like you don't understand a simple thing and become overwhelmed of any normal things and live can't understand that people can do normal things and live their lives. I suffer that straigt away when I wake up and whole day. I was prescriped a big amount of oxazepam for panic attacks 2021 (My dog had to put to sleep at the age of 16)
and doctor didn't warn me. Year ago I taperd it and brain lock was bad and i had terrible panic attacks and withdrawl symptons. Another doctor did crossover from oxazepam to diazepam and I got reliefe to my brain lock and could sleep and got home from hospital. Agoraphobia went away (normally don't have it) and I could function, I wasn't anymore in panic all the time with brain lock and insomnia etc. I could exercise and have a good feeling first time after my dog had to put to sleep. I could cook and ne with my partner and mom and other people. But after few weeks of tapering diazepam too quickly I lost my sleep, depression came back, agoraphobia, depersonalization and derealisation. After three weeks of suffering, puking, not sleep barely at all I took overdose and my partner found me uncounsious after four hours and I got to hospital. After a week I got home very sick. Doctor rised diazepam to 12 mg and I tapered it 1 mg per week in awful feeling. At 5 mg I went to hospital and they tapered me to zero straight and I had terrible panic attacks and brain lock still. After month rude doctor said I couldn't have any withdrawl symptons. I got home very sick. My new doctor prescriped me klonopin to get me relief for a while and then taper but it didn't help. Brain lock stays and that same panic and I am isolated a lot because I tell my hell all the time to my loved ones. I am just like same when I was in withdwal symptons of oxazepam year ago (and then diazepam got me to life back for few weeks, and I got to eat breakfast and drink coffee in peace and planning normal day, I thought now I will get off of this drug, had just few physical withdrawl symptons) and me and my mom said to my doctor that I think I suffer withdrawl even don't taper and want to taper with Ashton manual and she was willing to read it and we started week ago. I feel awful and brain lock is stopping me to function normally. My brain was scanned in the mri few months after my overdose and it was fine. Now my doctor thinks that I have tolerance and that's why klonopin didn't help. I want my healthy, active, good life back and I can't take a day anymore with this brain lock. I need to get this drug off of me nyt I don't know I will survive because I am in the beginning of long tapering and my brain is making me crazy. I can't be with my partner because I am on constant panic and not myself. I feel I will koske everything and I would have a good life, have always been grateful of all the good in my life. Can someone relate to that thing that feels like brain doesn't function normally? Please if someone can read this and answer and maybe give some hope that the lock can be part of the long benzo use and tapering. Thank you.
If you want to stop taking ur medical drugs without problems... Then take smaller dose some days ...then take smaller dose than before ...then take very very smaller dose than before ... Then stop using it 1 month ... 1st month it was hard cz of withdrawal symptoms... After 1month you fully releieved...

Switching off all the neurotransmitters for tons of other functions and causing a plethora of agony in withdrawal, lights to emotions on, agony, flicker flicker fail, zed out whole system. Seriously systematic failures everywhere and a person's trapped in that body.

Watching this after I took Benzos from a friend, it literally did nothing to me, how does it work for you guys? I feel like this stuff doesn’t work on me, Caffeine makes me sleepy, weed doesn’t make me tired or doesn’t give me any affects and now, benzos don’t affect me
it sounds like the pill gives you the effect of exercising without exercise
It kinda does talking out of experience, but without the exhaustion and you don't feel pain or something.

However, I’ve been using prescribed diazepam (Valium) and alprazolam (Xanax) for 11 years now after a severe and life altering spinal injury, but now I’m a fvcking mess!
I have fits of rage, long periods of deep dark depression and anxiety so bad I can’t leave the house unless I need pet food, but I’m the first to admit these meds aren’t fun after any longer than a few days, but 11 (eleven) years it’s ruined me and my once awesome life I really enjoyed.
Please don’t take this comment lightly, it’s truly that bad after only a few days then that’s it, you’re now a victim and dependant on a drug you either chose or was prescribed like me.
Please be safe out there anyone taking these meds for fun, I’m cool with that, but look out for one another, don’t let a homie or someone you love get to how I feel every waking naked sleeping moment….

It's really difficult to say for sure because it's not clear at this point if GABA supplements are even having an effect on the nervous system. I would speculate that the GABA supplement would NOT negate the effects of the benzodiazepine. If there were a synergistic effect, it's hard to say at this point if it would be due to anything more than a placebo effect + the effects of the benzodiazepine. Binds to the same receptors. But in a diffrent way tho. Thats why alcohol an benzos feel completely diffrent when you take them. They work diffrently but similliar at the same time.

"They are thought to work the same." They are believed to work by the gamma..." Thought, believed, the truth is they dont work. The longer your on them though the more risky it is to get off them.

benzos do work pretty well, but are pretty risky. you probably already know this, but if you just started taking them, do not believe doctors who say benzos arent addictive. they are harder to quit than opiates

I don't know about shortening it mate. Unfortunately being in the same boat myself the only thing I know shortens withdrawal is time plus Flumazenil.
That is an antagonist of the benzo receptor and can help a process called recoupling.
If only... I could access that. It could be life saving for millions of miserable people
I think meditation and mindfulness can help some people alleviate intense withdrawals.
ASMR is good too but not everyone responds and some think it's weird but I don't care now. Most haven't experienced the misery and good for them as I wouldn't wish it on anyone.

Not much, but the effects of the two drugs would negate each other to an extent that would dependent on the dose of each of the drugs. The caffeine would counteract some of the sedating and anxiolytic effects of the benzos, and the benzos would counteract some of the stimulating and anxiogenic effects of the caffeine.

Ppl take this for anxiety and sleep issue? Then suffer hell for years to come? Sounds evil. What people should do is playing something calming to fall asleep (mine is science or natural science programs to listen to. Stimulating at first but over time became familiar and sleep inducing, more effective than music). Or whatever natural, you body will be tired and sleep. What the hell!

No. What kind of doctor prescribes both? How would it be safe? Benzodiazepines use GabaA and Glutamate receptors to effect the CNS and PNS, Opiates depress the CNS. You have two agents that are depressing the CNS... How would this be safe. They put out a public awareness of showing that benzos with opiates are deadly
