Stuck injector likely. Noid light should help rule out whether or not the computer is commanding the injector to open. Looks like it fills once the fuel line primes so likely a leaking injector IMO

vacuum lines, there is a tool you can buy to set the butterflies and vac lines for a holley carb

WRONG.... This is a TB, NOT a carb... That is a leaky or stuck injector.

That is a stuck injector that is losing its load during the pre-crank prime portion of the start up sequence.
