If primary antibody has the affinity to bind to the well, then antigen vil also have the affinity to bind to the well n there is a chance of binding of secondary antibody to the antigen n gives us a colour in the end.

I have one question why we skip washing step in direct Elisa because we are using enzyme linked antibody and in the next step have to add substrate , substrate because product by enzyme or if antibody is not linked to particular analyte but we have enzyme linked antibody and substrate in solution so is there any chance of colour production. I am so confused please explain
Sir as you had told antibody- antibody don't bind to each other on time 12:56 in video. but in previous video of yours for topic Western blotting you told that antibody binds to antibody Fc region and shows color in solution.
But the Fc region is already attached to well or may be both antibodies are not similar to bind each other ... As per my understanding i think that might be the reason ab doesn't bind to ab

well, your question is very good but my point of view the answer is that although Ag Ab bind together so secondary Ab also follow the same lock and key model it does not follow any other method to bind with Ag, lock and key model does not say that one Ag Ab phenomenon always bind, does not matter how many Ag or Ab bind. Hope u got it
In the case of sandwich ELISA, we have a secondary antibody that can interact with a particular region of primary antibody then why after second washing and adding substrate it will not show color?

Affinity to bind well but it is fixed in well
Elisa plate or well is coated with blocking agent that prevents further binding or unnecessary binding
