  • Petri dishes are something we all have to work with. This said, they can be a real pain and because of this a lot of growers put making them off for a while. The No Pour Method makes fixing up small scale or on demand batches of petri dishes super easy. These containers make no pour method easy and possible, and add other benefits on top of this!

    I immediately went to look for those petri dishes as soon as I saw how large they were in your hand, and that they're PP 😂
    I scoured the website you linked and found a couple other items that mycologists might find useful, but rather than petri dishes these would be useful as culture slants: item# 2924B01CLR and item# 314940 (the latter of which I imagine will be the better option, but I'm buying both to see). They're both clear PP tubes with hinged snap-on lids. I'm hoping that with Parafilm, the snap-on hinged lid design won't pose any issues.
    These are all cheap as dirt -- *MUCH* cheaper than proper lab supplies... so I'm happy to try these out 😁

    You can also drill a hole in the lid and add an injector port to them. Great for starting a LC.

    I like how them containers are sealed you know what’s in side is contam free
    I’ve been doing the about the same thing using a short deli container I get from local grocery store or Menards make sure there’s #5 on bottom container poke hole in lid cover with 20mm filter patch or micropore tape and go at it

    Thanks for sharing your knowledge on these instructional videos! Super helpful. A couple quick questions... I wanted to try out the Petri Dish replacements you recommended but the page linked to Berlin Packaging isn't working for me (it turns white and fails to render properly even when I navigate through their menu UI). You mentioned having other sources for these dishes. Could you share an alternate link? Also how do these jars allow gas exchange?

    Thanks for this video! i love no pour techniques and yours is pretty cool.
    i currently use larger reusable screw on lid dishes, but i want to migrate to those smaller ones. Have you found any of those smaller ones that has more transparent plastic?

    Great Video! I've been using those same cups for a while and Love them! Now, I mix a batch of Agar and pour all the cups after. Any reason you mix them in each individual cup? Thanks!

      Thanks! Yeah, I use a Still Air Box, so I don't like to pour them in the SAB. I like that after the PC is done, I can just tighten the lids and I'm done.

    When you go to pressure cook them, do you put them in a jar or a bag? Or do you just put them into the PC as is?

    So we should let it build a vacuum when cooking? What about leaving the cap loose slightly then tightening after it sets so no suction when opening again?
    I was expecting you to say leave the caps loose like liquid culture and was surprised to see the vacuum so that’s why I ask.

      I don't leave my LC lids loose, and no I don't let these stay loose either. As they cool they can suck in contaminates.

    Is the toothpick necessary if not for long term storage? I thought it was for nutrients after the agar is used up, thanks.

      Indeed, the toothpick is for long term storage usually in slants as mycelium will colonize the wood and outlasts agar from what I've read. I guess this could be a good longterm agar dish solution??? But why

      I'm thinking you could use them like that, but I would want long term testing done first before I switched from slants. I also prefer storing the slant shape over the dish.

    Have you experimented with putting a syringe filter on the lids for air exchange? Or is it not necessary?

    I put the food coloring in the water. It seems to be more consistent across multiple pours.

    I believe PastyWhite started this years ago on the Shroomery. It's easier to just mix, pour, clave.

      You can certainly do it that way. If you want to make up only one recipe I think that's not a terrible way to go.

    How do you keep it ventilated after inoculation? Do you keep the cap loosely covered? Do you still use parafilm with this method?

      No parafilm, no need to vent. Lid is not kept loose.

      @@JinTalzeus There is some air in the container once you've worked with it in front of the flow hood, but mushrooms do not need as much air exchange as we often think unless we want rapid growth of a lot of biomass. Interestingly enough as well, many fungi can pull oxygen from water molecules and use that for their metabolic needs. It's slow, but how they often grow deep in a log without access to fresh air from outside the log.

    What's the contam rate like? When opening, the vacuum could pull contaminates in? I guess not so much of a problem if you got a flow hood.

      I assume you could leave the lid cracked slightly and let them cool in the pressure cooker overnight, when you release the seal on the pressure cooker make sure to then tighten all the lids.
      The inside of the PC is sterile until you pop the lid, so letting them cool in there should be fine contam wise even with the lid cracked

    Mictopose makes PP 5 autoclavable 90 mm x 15 mm Petri dishes that you can buy in cases of 500 or smaller amounts

    So I've scrolled through vid like 10x and can't seem to find the recipe amounts for these cups but I think I remember them
    1/4 tsp agar 1/4 tsp lme and 20ml water
    Am I remembering correctly?

    Here is a question: Petri dishes for mycology typically have vents for gas exchange. These twist top containers do not (as far as I can tell). Isn't that a substantial con?

      Tarrison is probably the best but holy $$$. The ones you buy at home depot or lowes are a lot cheaper and are pretty decent.

    Soooo what's the toothpick in each dish for??? He said "you can use a toothpick or sawdust", but the only thing I know sawdust is used for is as a fruiting substrate, not in agar petri dishes...
    Can anyone please elaborate? I'd appreciate it.

    It seems so tedious to make those small measurements and inject water into each dish separately. Would it not be easier to just make up a batch, mix once and then pour a little in each dish then pc them?

      You can certainly do that if you so desire, but I don't find that easier especially when I'm making up dishes of different recipes in the same batch.

    I tried this. Didn't go well for me. I used the ones from Amazon before he changed the link. My problem was how deformed they got in the pc. The top and bottom are so concave that the agar is touching the lid in many cases. Also a lot of them leaked out and they were sucking in air as they cooled. I'll still try to use the ones that look OK but I'm not a fan of how it came out for me. Things that I could have done to mitigate this:
    -Put a layer of quart jars on the bottom of PC. I noticed the bottom layer of the no pour dishes were the most deformed.
    -not close the lids as tight on the jars before pc. Should they be tight?
    - I released the pressure after it was down to 5 psi. Probably should have just let it cool in the pc. This could even have been my biggest mistake.
    Are the toothpicks really needed? When the bottom of the jar warps the toothpick comes out of the agar. I will try again with the above tweaks, the new jars, and update. I'm really attracted to being able to re use these, also being able to sterilize them and putting an injection port seems really smart.

    Are petri dishes suddenly not clavable for some reason? The ONLY difference I'm seeing is no tape.

      plastic, disposable dishes are indeed not autoclavable. The big difference is in no pour and no tape.

      I've done that a fair amount as well, not a bad way to go. Used to sterilize a lot of batches in media bottles and then melt and pour after the fact.

      As long as you are clean and sterile it is easy peasy. Tissue onto agar or into liquid culture is just amazing. It works so fast.

    K cups or ketchup cups are cheaper than this and just as effective. At least you can get the lid off. Can you make videos with teks you came up with, instead of using other peoples. Very sad after all these years you make very little improvements on your own to share.
