For a sec, I thought the challenge was to do all 4 requirements in a single run (speed run a gold stake run, without jokers and get e20/e40) which...as insane as it would be, I figured "it's DrSpectred, of course he got away with it"

e20 jokerless gold stake sounds horrifying

e20 jokerless on a non-plasma deck is theoretically impossible. In his recent jokerless high score run he got into the hundred billions (e11) and with perfect luck a similar run could get into the trillions (e12). But planet scaling without jokers just isn't fast enough to get to e20 from there (ten million times what would be considered a run with perfect luck)

should have opened the emperor at 3:52:50 while in the arcana packs, this way it blocks 5 cards and gives you a bigger shot at death

In this position, we're holding Fool and Justice to glass the Red Seal card. Using Emperor will only generate one card, with 1 in 14 chance to be a Death. If we wait to use Emperor after the Fool and Justice, we can spawn two cards from the Emperor, in which case we have slightly better than 2 in 21 chance of getting a Death card (higher than 1 in 14).

Let's say hypothetically someone managed the High Score requirement before Ante 8, and then somehow lost before Endless Mode - would that be grounds for expulsion?

High Score doesn't require completion!

what if get a negative copy of every planet card with Perkeo, and then open planet card buster, its give 1 black hole every time or game crush?

All cards in the game have a default that the game resorts to if it cannot make one normally. Planets are Earth, Jokers are Jimbo (even if the rarity would be wrong, i.e. you've run out of Uncommons but you still have open Commons), Tarots are Strength, and... I'm blanking on the default Spectral since it's so incredibly rare.

eh I have all these nice
gold stake on like 3 or 4 decks
finished all the challenges including the jokerless
easily have like less than 3 minutes run, runs on plasma deck are so easy
and E69 on plasma deck

You have to do it all consecutively. And you have a 3 minute run on a random seed? That's exceptional luck

That's part of why the requirements are to do them consecutively. If you spam Full Skip runs on Plasma, you'll lose 99% of the time, but the one time you get Bull, you'll win in less than 3 minutes without much skill needed (in the sense that there aren't any decisions to make).
