Telomerase Replication in Eukaryotes | End Replication

In this video we have discussed the Telomerase replication in Eukaryotes. This happens when there is end replication problem in eukaryotes after removing the last primer of lagging strand during DNA Replication process.
The telomerase replication is driven by Telomerase enzyme .
Telomerase, also called terminal transferase,[1] is a ribonucleoprotein that adds a species-dependent telomere repeat sequence to the 3' end of telomeres. A telomere is a region of repetitive sequences at each end of eukaryotic chromosomes in most eukaryotes. Telomeres protect the end of the chromosome from DNA damage or from fusion with neighbouring chromosomes.
Telomerase is a reverse transcriptase enzyme that carries its own RNA molecule which is used as a template when it elongates telomeres. (Beyond unicellular organisms) Telomerase is active in normal stem cells and most cancer cells, but is normally absent from, or at very low levels in, most somatic cells.

@@hussainbiology Q- Telomerase adds 6 nucleotide repeats to the ends of chromosomes called telomeres. The telomeric repeat sequence of tetrahymena is TTGGGG. The telomeric repeat sequence of yeast is GTGTGT. Propagation of terahymena chromosomes in year will result in extension as follows
a. TTGGGG TTGGGG TTGGGG
b. GTGTGT GTGTGT GTGTGT
c. GTGTGT TTGGGG GTGTGT
d. TTGGGG GTGTGT TTGGGG

To my understanding, the overhang flaps are essential for the formation of t-loops and are not removed by Fen1. T-loops seal the ends of linear chromosomes and bind TRF proteins. It's a way of protecting the telomeres that would be otherwise interpreted as damaged DNA. I think the overhangs are actually elongated by an 5'-3' exonuclease for this purpose, and an overhang flap is produced on the leading strand end as well.

correct. before fen 1 removes it, the overhang is looped to form t-loop and proteins trf 1 & 2, pot1, tpp1 and rap 1 forms a protein complex called sheltrin that binds on and protects the t-loop structure.

Thank you!! Excellent❤🔥 But a little note, we learned at the uni that at each fork we have a leading and lagging strand. The primase puts the 1st primer of a given strand at the midway of the replication eye, so each DNA strand is leading from a side and lagging from the other. Idk if I conveyed the idea well. But overall, the video is marvellous; it's only now that I understand the telomerase concept.👍👏❤

FEN1 does not cut the overhang DNA telomere sequence.. Instead there are some telomere binding protein like POT1 are recruited which protects the telomere end as the single strand of telomere can send signal for DNA repair. In alternative way the overhanging telomere sequence can also form a t-loop like structure.
