Protein Quantification using BCA method

The Bicinchoninic acid (BCA) protein assay is a colorimetric detection and quantification of total protein. This is based on the reduction of Cu+2 to Cu+1 by protein peptide bond and amino acid in alkaline medium known as the biuret reaction. Further, the chelation of two molecules of BCA with one Cu+1 (cuprous ion) gives water soluble complex of purple-color which exhibits a strong absorbance at 562 nm. Increased absorbance represent the increased amount of protein in sample. Bovine serum albumin is used to prepare the standard.

Another effort to make a video on BCA protein quantification method.
