Hey Todd, No, you should never fully plug the roof vent. For a smoke test to be effective the smoke must flow freely through the system. In some cases it is acceptable to partially restrict the vent to create a small amount of pressure but generally this is not required.
Hey Brian, That is not a bad precaution but any embers from the fuse generally do not survive beyond the blowers squirrel cage. Also by turning the blower on and establishing a draft, the piping, especially near the blower, is flushed with fresh (non explosive) air. Turning the blower on first is also a good way to verify there are no restrictions in the system.
Those vents have to be install upright, not horizontally.

Should be vented through the roof. If the the drain backs up the insta- vent can become clogged and allow the trap to syphon

I thought you were suppose to tin the machine on until the flame turns to smoke to avoid explosion
