You can do both... the 1290R has a 2-way quickshifter clutch so in ROAD mode you can use it.
But that 141 Nm torque is available from the first millisecond you touch the twistgrip.
Due to it's design it will even do power wheelies in 5th gear as you put it on SPORT mode and disable MTC.
