Actually Margaret who up to this day never had seen a man has fallen for Luffy without being aware of it. But the rules of Amazon Lily demand Luffys death, so she is conflicted. And it is easy to explain why the village people did not recognize a Luffy as a man: Only Kuja PIRATES have left the island and therefore met men - navy members ore people they raided. P.S. Granny Nyon is a former snake princes as well, but the title is not inherited, so she is not related to Hancock. Once the title gets available either by resignation or death of the actual snake princess, the strongest Kuja advances on the throne - so Hancock is obviously not a pushover.
