I am trying to just do Packet, and at 3:51 you are showing VARA and where I live we only have packet. I don't see anything in Winlink Express that looks like what you have. (that sound card)
Sorry about the delayed response, UA-cam has been having trouble providing notifications about comments that have been posted to our UA-cam channel. I have reported it to them. In Winlink Express in the top menu bar, you will see Open Session and then there is a drop down selection that you can pick. In my case I picked Vara FM Winlink. Once that is selected you can click on Open Session. The screen that is being displayed is under the Settings menu in Vara FM then select Soundcard.

I recommend setting the squelch level to 0 unless you have a lot of noise. If you have a lot of noise try it set to 1 but no higher. The only other radio settings were the frequency. That will be determined by the Winlink gateway that you are attempting to connect to. In my case it was 145.030 simplex with no offset or PL Tone. The volume knob setting on the radio will be determine during your tuning setup. As you saw in the video you have to play with the volume knob settings as it has an effect on the Drive Level, S/Noise, and VU.
