Yes, solvent itself will be considered as a blank in one trial

You can use different solvents to extract your compounds. And also can check their pH. Preferably a weak acid, neutral or strong acid solvents you can use to check their growth . Also take in different concentrations. For temperature you can do in two ways, you can alter the extraction temperature or incubation temp.

Hi Sir, may I know during incubation of well diffused petri plate well it be okay if I invertedly position the plates?Will the inoculated extracts would stay in tact?
In my experience the solutions added in to the wells get absorbed and then you can invert the plates within few hours.

You can dry them for few seconds and immediately put. The more time more chances of contamination. Moreover as you add drop wise the disc absorbs the extract so it won't be a issue

Liquid extract is used. If it's solid redissolve in suitable solvents.

Antibacterial is how efficiently extracts kill bacteria, antitumor is how efficiently tumor/cancer cells are killed by plant extract

May I also ask this one, Sir. Can we use Whatman No.1 filter paper as an alternative for the antibiotic assay disc? If possible, does it affect the ZOI of the assay solution? Thank you so much!
I don't think so in papers such details for positive control will be given. Anyway u can browse them. If you're using well method directly you can pipette the samples control into well. Disc method requires the disc as it has a known conc. of equally coated drugs which cannot be compensated by filter papers.

Sure. Make the disks and the autoclave them.

No whatmann doesn't serve the purpose. You can try as a trial and error and check. The one I used was purchased not made. So I don't know the make. You can buy it as sterile disc's from internet. Or you can use even without that by well method or broth method watch that video too.

Hi I have two questions related to the topic (+ I just want to say thank you so much, these videos are exceedingly helpful) :
1. if I am testing an antifungal activity of a substance (garbage enzyme from citrus peels to be specific), can I use well diffusion method?
2. what is the most reccomended method for inoculating the fungi? Is it the same method shown in the video (using liquid spore culture first, then the liquid is spread into the plates), or can I simply transfer a portion of a culture inoculated in a plate using a scalpel? Or is there other beter method?
I am using Botrytis cinerea as the fungi btw. So it's antifungal activity of garbage enzyme from citrus peel againts Botrytis cinerea
From what I can understand so far (by researching the topic on the internet), the standardized/more reccomended method is using broth dillution, but I am also under the impression that well diffusion can also work to test antifungal activity againts Botrytis cinerea from a study I had found titled "A Comparative Study of Antimicrobial and Antioxidant Activities of Plant Essential Oils and Extracts as Candidate Ingredients for Edible Coatings to Control Decay in ‘Wonderful’ Pomegranate"

Its based on volume thats all. Not quantitative

Yes you are right, It was just for demonstration to show the audience. I haven't used any standard antimicrobial drugs here.

No they are just plain discs which will absorb ur material or samples. You get antibiotic coated disc which we use as std. If you don't get that you can buy a antibiotic from chemist and dissolve it and use that

Dear sir, please help! I am conducting the antibacterial assay of my plant sample following the well diffusion method. But I have faced with a problem. Should I filter my plant sample with the help of 0.2 micron filter inside laminar flow cabinet before adding the samples into the wells. I found that when I filtered my sample, the concentration as well as the volume of my prepared sample also get decreased. I have also gone through severe literatures of my field but found no paper mentioning the use of 0.2 micron filter. So is it necessary to do so or I can skip this step?
You should not filter the extract . Add directly.

Just mention that there are no papers who have followed that filtration step.
And even if you filter it, after filtering make up by adding the same solvent. Solvent need not be filtered since your using that as a control in a well. So make up the concentration using the same solvent

Dmso is a solvent. Like water , methanol ethanol it is also useful to make extracts.

Ok, then venom is usually too toxic and concentrated. You can dilute it with water. Say for eg. 50ul in 450ul water and consider this as your stock. From this pipette different concentrations may be 25, 50, 75, 100 or 50, 100, 150, 200 etc
