Thank you so much for this. I think the calculation was wrong at the point of 800microliters of the 1mg/mL antibiotic solution. it should be 80microlitres in 19.92 mL to obtain 4mcg/mL. The calculation would however be correct if it were a 0.1mg/mL antibiotic solution. But I give kudos to this simplified and practical explanation. Great job.
On a second look, I think the concentration has been brought to what was actually labelled by diluting with 90 mL of agar. We can then say the 4mcg/mL in tube was actually 40mcg/mL and 10 mL of that was diluted with 90 mL of agar, to give 4mcg/mL. hence the agar dilution as its name implies
Hi there, please help! I am conducting the antibacterial assay of my plant sample following the well diffusion method. But I have faced with a problem. Should I filter my plant sample with the help of 0.2 micron filter inside laminar flow cabinet before adding the samples into the wells. I found that when I filtered my sample, the concentration as well as the volume of my prepared sample also get decreased. I have also gone through severe literatures of my field but found no paper mentioning the use of 0.2 micron filter. So is it necessary to do so or I can skip this step?
First of the tube or container is labelled as 4 mcg/ml and others with 10 ml of distilled water are marked as 2, 1, 0.5 mcg and so on. To the first tube you add 19.2 ml of sterile distilled water and 800 ul of antibiotic stock this will give you 20 ml of first dilution ( 4mcg/ml) now serially transfer 10 ml to prepare the next dilutions

Do u happen to know , how to like use phyto oils in mic . They probably wouldnt diffuse in the agar surface
How many ml of agar was prepared for each plate and what is the volume of the drug diluted in it?
