Autoclaving Tutorial

One of the miscellaneous tasks you are responsible for is autoclaving pipet tips, pasture pipets, and Eppendorf tubes. You may only need to do this about once a week, depending on the amount of experiments you and others will be doing in the lab.
Tips:
- Although you won't be doing this so often, try to keep up with the empty pipet boxes as they can add up quickly during busy weeks.
- When adding the autoclave tape, fold one of the corners so that it is easier to take off when it becomes empty
- When you start using a newly-autoclaved pipet box, move the piece of tape to the top of the box so that you and everyone else knows that the tips are clean.
- You can stack up the pipet boxes over the brim of the autoclave container so long as they don't fall over.
- Whenever you are autoclaving empty glassware, cover the opening with foil and also use autoclave tape. These can be stored for a long time so long as the opening remains covered.
- For dry objects, use the gravity setting on the autoclave. 10-15 minutes is enough time.
-For liquids, 15-20 minutes is enough time.
-For liquids in bottles, DO NOT TIGHTEN THE CAP ALL THE WAY as this can cause the bottle to explode under the pressure. Screw on the cap so that there is still play and use a long strip of autoclave tape to link the cap with the side of the bottle.
- Ensure the autoclave door is very tight before starting any run; otherwise the whole room will turn into a sauna. Don't ask how I know this.
