  • Drug Distribution | Volume of distribution | pharmacokinetics | distribution of drug
    A simple explanation about drug distribution and Volume of distribution with two-compartment models.
    Vd = X (mg)/Pc (mg/L)
    high Vd - Plasma concentration is low and Tissue accumulation is high
    Low Vd- Plasma concentration is high And High plasma protein binding.
    Volume of Distribution mean drug Distribution within the body and whole body has some volumes but exactly we can't measure the volume of individual body thus we used a term Apparent volume of Distribution
    Methameicaly Vd = X/PC
    X= Amount of unchanged drug present on body in mg (iv administered dose)
    Pc= Plasma conc. In mg/L
    Means a drug is distributed in Central compartments (Blood) and Tissue compartments.
    And one thing should know, the drug, which is present on blood, acts therapeutically. And the drug which present on Tissue may cause tissue toxicity.
    1. High Vd = means Tissue accumulation
    2. Low Vd = mean drug retains on blood
    For example
    Let us think Total body volume is 10 L and Plasma vol. 1 L
    And if we administered 10 mg of drug by iv routes means X= 10 mg and initially at time 0 all the drug present on blood/plasma so
    Vd= 10/ 10 = 1 L
    And later at time t 9 mg drug is distributed in tissue and only 1 mg drug is retained on blood so
    Vd= 10/1 = 10 L

    Ionic drug can be dissolve easily but not cross cell membrane and unionic drug not dissociate easily but they are cross the cell membrane.. So we make a drug in such way that before time of dissolution they are in ionic form but at the time of absorption it convort to non ionic form

      Yes, therefore we make salt for of most of the drug for oral formulation

    Sir Unionised drug k adissolution rate kam hota hai to vo kese metabolism hoke membrane ko cross kar sakate hai

      Do different Chij puchh rhe
      1 Union drug Easley cross kr skte membrane ko metabolism ka koi roll ni
      2. Metabolism hone ke bad wo ions ni banate, chemical nature chng hoga
      3. Drug orally lene k bad, wo phle dissolve ho ga fir membrane tk phunchega, fir us drug ki individuals lipid solubility ke base pe wo absorb hoga, fir metabolism ke lye jayega
