I guess eyeglow in Undertale signifies that they're on their strong level. Asgore is basically always on his strong level, except on genocide and pacifist, because then he's either afraid of you, or he doesn't have time to fight you. Undyne's eye shines strongly like a laser because she's super determined to kill you and therefore she becomes super strong, but only on genocide. Mettaton's eye glows becaue he's at his strongest, because it's genocide, too. Sans though, his eye only glows when he's using his toughest attack, and that *only* on genocide, because he's too lazy to keep on glowing his eye all the time, and he isn't that strong. Or he's just effective, and he uses it to a minimum to spare his strength.
