    Hi, I am a great fan of yours as you are doing an awesome job and your videos really help me a lot. But still I have to make this comment so that a misunderstanding does not take place. In the video at 8:28 you said by mistake that ATP converts luciferin into luciferase which is not true. Rather ATP converts luciferin into oxyluciferin that gives luminescence in presence of luciferase enzyme. I know you must have a clear idea about that and you probably had a slip of tongue (because later you only said from luciferin, light is produced with the help of luciferase). So, if you could kindly mention the correct information in a pinned comment or in the description then I think no further confusion will take place. Cheers! And keep teaching us!

      @@MuhammadIslamag adenosine phosphosulfate, as it is a substrate and the enzyme that converts these ppi and substrate into ATP is ATP sulfurylase.

    Shomu da very informative video. There is one slight mistake : APS is adenosine 5' phosphosulphate. I have one doubt what is the use of adenosine alpha thiophosphate ?

    Thank you Suman sir. It was nicely explained. In this regard I would like to clarify that here APS is Adenosine-5'-phosphosulfate, not ammonium persulfate. Hence it reacts with PPi to produce ATP. However your video is very helpful.

    I have query : In a well we have multiple fragments (having varied sequence), then how can we detect the sequencing because at the same time nucleotides are added at the multiple fragments ?

    APS doesnt stands for ammonium per sulphate but it stands for adenosine phospho sulphate. still your lectures are really helpful

    Well explained though i have a question: in other websites I saw that there is a step where we do PCR that you are not mentioning, when do we do the PCR? before the fragmentation of the DNA sequence of after it ?

    What determines the sequence of the fragments after the genomic DNA is fragmented and attached with the adapters. The original sequence is maintained by what means? The fragments may not bind to the beads sequentially,.. thanks for the videos

    Arent adaptors added to both ends of the DNA?? And how can you break a double stranded DNA to a single stranded DNA?? and upon breaking the strand, how will you seperate the template and the complementary strand??
    please answer this at the earliest

    454 sequencing and pyrosequencing are different? I was always told they are the same.

      I think Roche 454 is a comercial name. The biochemical name is pyrosequencing.

      454 method is a general procedure to perform pyrosequencing as well as ion torrent sequencing.. ROCHE 454 is a kind of emulsion PCR technique.
