40 mpa is target strength decide for perticular characteristic strenght .. if there happens to be bad workmanship , high w/c ratio will least get 20 mpa
which equipment is used to compact the concrete filled in moulds? what is the ideal weight/area in kg/cm^2 for M45, M30, M15 grade concrete respectively? How much load is being applied per cm to the concrete cubes surface if a manual, lever arm operated Compression Testing machine is used? In automatic Compression Testing Machine, as mentioned by you, the load transfer rate is 140 tonnes per cm. Please correct me if I am wrong. Also if the mixture is taken according to 3 moulds quantitatively, then if the readings aren't 15 plus or minus of each other, the whole mixing process has to be done again?

In real case is sample to be collected from Construction Site during its construction (i.e slab)or 1st we prepare in lab then after we get results we start in real construction filed? simply testing is done in lab before construction or during the day of construction sample is taken to lab?

TILAK SURYA VARMA PASUPULETI nothing. The standard says MINIMUM of 35 tamps per layer. so more won't affect the strength but less will.
