Yes! I sometimes use multiple strands of yarn at the same time, even without twisting them.

can the dowel be placed before starting the weave?
I've never tried that before, I think there's a few potential issues, one being that it's going to make your warp a lot tighter so you'd have to plan for that. I also think that would work better on a peg style loom where the pegs are perpendicular to the loom. Either way though the loops will always be slightly too long for the dowel anyway since the warp is going around a peg or notch, so in my opinion it makes more sense to add it after.
