    I probably should've mentioned that a larger clinical trial is being conducted in Singapore using AKG and looking at more measurements (Prof Brian Kennedy) so hopefully we will get more data soon!

      Given that Prof Kennedy is the Chief Science Officer of Rejuvant, there will likely be some inherent bias in that trial as well.

      @@Holypikemanz Its AKG with Arginine added, which is a Nitrogen booster in the body, which could be good or bad depending on what type of metabolic dysfunction you are suffering.
      Calcium-AKG or Creatine-AKG seem to be better. Get your NO2 boost from food/diet or take arginine in isolation.

      The data is extremely biased, which puts it well down on my list.
      The fountain of youth has always been a great sales ploy and now more than ever so ethics are paramount.
      Other compounds like Fisetin and Rapamycin looking better to me.

    If a supplement makes me 8 years younger by objective measures, in a about as many months of taking it: I don't care if it's caused by placebo, magic fairies, or its ingredients. I would be interested in seeing other markers tested though.

      Yes - A test like this would need to be done by independent third parties - using a bunch of the top age biomarkers / epigenetic clock techniques.

      @@thaidomain I have studied the benefits of AKG and do take it daily. I always find the source of any publication before I read…if the participating PhD’s have any visible ties I don’t bother reading, but their ties are often hard to discover 😞

      Science would have made it nowhere without financial gain. You know how much money you lose if you don't commercialise? It costs millions sometimes billions to bring a drug to market, for example. Without a product you are throwing away your money, basically. Crowdfunding studies would obviously be an alternative approach.

    I take AAKG, but I use it more as a once daily supplement. Except for days I am exercising heavily, then I take 2 to 4 scoops throughout the day. I can tell the difference when I run out. For me it is noticeable.

      It's difficult to attribute this to AKG specifically; it might simply be the vasodilator effect of Arginine.

      There was speculation as early as the 60s and 70s about this exact thing. I obviously don't think there is anything to it. But books were written in the new age fu-fu section of the bookstore about it.

    It helps correct the amino acid imbalance which causes cascading failures in aging, and contributes to the dysregulated integrated stress response that decreases proper protein production, leading to frailty.
    Calcium alpha ketoglutarate corrects secondary hyperparathyroidism, keeping calcium in the bones, which safeguards arteries and organs from being exposed to it. Helps with phosphorus metabolism and homeostasis, and also helps stop disruptions of circadian mediated autophagy.
    I've messed with a lot of different AKG formulations, from Calcium-AKG to Creatine-AKG to DiLeucine-AKG. Works better with Serine or Glycine mixed in.

      Awesome! So, you mean Ca-AKG will by itself help deposit Ca in bones and does not need the help of D3+K2 & Mg ? Also, I suffer from insomnia. I tried melatonin 300mcg and failed to sleep. I had taken xanax type of pills earlier but don't wanna go back to them. Do you think Ca-AKG can help with insomnia? And, did you find any noticeable anti-ageing benefits from this supplements? I heard many reversed their hair greying. Would love to know your experience.

      @@aliengod2039 You'll always need basic nutrition. It just counteracts Calcium and Phosphorus metabolism dysfunction in aging.

    Someone should do a randomized placebo controlled trial using Arginine Alpha Ketoglutarate which is cheaper but has shown effectiveness in antiaging in other species

    It's hard to trust a study sponsored by the company that makes the supplement especially when they're charging $150 for a one month supply. Other companies can make a A-AKG sustained release supplement two month supply for $50.

And it doesn't have to be some chemical concoction, you can take it right away too and have impressive results on skin, digestive health and increased cell apoptosis and anti inflammatory.     Interesting to note that the lines on the delta graph basically have two different slopes. One group saw moderate improvement the other saw much more pronounced improvement. Would be very interesting to see if a relevant variable could be identified to distinguish them.

      How much attention the clinical staff paid I am guessing. Some doctors throw pills at you and are surprised you come back. Some insist you come back, ask a lot of questions, have their staff ask you questions say nice things. Even CALL (the nurse does) if you miss and appointment or don't make one at the right time for a refill! Imagine that! Given that elderly patients often feel alone or ignored by family and/or friends that counts for a LOT.

    With all the shortcomings considered (not placebo controled and non assement of some functional data being more serious, the others - I don't see them as important), I think this is an important study as it points to something else than NAD+ boosters. Thank you for this post.

      There has been something other than NAD boosters for quite awhile. I would argue that inexpensive Glucosamine has more data behind it than NAD boosters and AKG. The Govt or some other org should fund RCTs of Glucosamine
      "After controlling for various factors -- such as participants' age, sex, smoking status and activity level -- the researchers found that taking glucosamine/chondroitin every day for a year or longer was associated with a 39 percent reduction in all-cause mortality.
      It was also linked to a 65 percent reduction in cardiovascular-related deaths."
      West Virginia University. "Glucosamine may reduce overall death rates as effectively as regular exercise, study suggests." ScienceDaily. ScienceDaily, 1 December 2020
      Research out of Tulane University also found a correlation with use and benefits
      "The researchers found that glucosamine use was associated with a 15% lower risk of total CVD events, and a 9% to 22% lower risk of CHD, stroke, and CVD death compared with no use.
      These favourable associations remained after taking account of traditional risk factors, including age, sex, weight (BMI), ethnicity, lifestyle, diet, medication and other supplement use.

      So I just take that for now.

      Steve Horvath already tested that NAD and NR booster. Both do not reduce the biological age at all. Resveratrol and Stem Cells do though. Stem Cell are not mentioned enough on this channel. Just stick the cytosine clock and use Ca-AKG and stem cells (or exosomes from stem cells)

    It would be really interesting to hear from anyone that says they did the any methylation time tests, took the product for any lenght of time (4 months, 6 months 7 months ) and then did another methylation clock test and found a decrease. I've done the Truage test myself - I'm 52 - and sadly my Truage test came back showing me as older with an age of 60 - so after reading about this I'm contemplating giving this a try for 4 or 6 months and then testing again to see if it actually does anything to the methylation test results - would love to hear from anyone else who has already done this "successfully" - I'd probably get calcium AKG supplement other than the brand this company is selling because their product is pretty expensive... would love to hear from anyone who's run age methylation tests and showed a decrease in "age" on the clock after using calcium akg

      @@enalo6261 hey I just saw your message about retesting - and I did a retested today. I didn't buy the Rejuvant brand - I bought a cheaper one (Prohealth) and I took it for about 4 months before stopping - I also took Prohealth NMN at the same time - so I dont know if there is really a difference between the version I took and Rejuvant (Rejuvant says their formula is coated to survive the stomach) or how much difference that makes. Anyway I redid the test today and i was pleasantly surprised. On the first test I tested as a 60 year old and I was 52.5 actual years. When I retested those numbers dropped - I'm 53.1 and tested at 58.2 or 5.1 years older. So the trend seems to be running in the right direction. If I were able to keep running it in this direction then at some point I could theoretically get it to where my epigenetic age is lower than my actual age. It's very interesting and I'm very surprised. I really didn't think it was going to do anything and thought I might just be even old epigentically because I have been having some health problems and not feeling all that good. I should also say that in the time between testing I went mostly vegan and lost about 20lbs. So I don't know if the supplement did anything or it was the vegan thing or a combination. I'm going to start the CaAKG again and will test again in 6 months. I can't say I really feel better in any way if you're wondering...

      has anyone else tried running these tests like me - super curious to hear from others. the idea is so intriguing - i wonder if I would have better results if I took their formula because it would be better absorbed and survive the stomach. Their formula included some calcium - why I don't know - but I did not take calcium. If I try again and do 6 full months instead of stopping after 4 would I have seen better numbers? if you wonder why I only took it for 4 months - it's because I started having some other problems where I wasn't feeling well - so I stopped all supplements for a while to see if maybe it was something I was taking (including the CaAKG) that was making me not feel so well... so many factors at play always with stuff like this hard to definitively say anything. Maybe the test would have shown a decline even if I hadn't done anything. who knows. The numbers look better but I don't feel confident that it was the CaAKG that did it - but I'm going to keep trying it. I would love to hear from others who are experimenting with this.

    Well done. I appreciate your approach of presenting the paper as objectively as possible, then providing logical critique. Your points were spot on, specifically the lack of control and the choice of methylation test. I am largely unfamiliar with attempts to connect methylation with aging, as I have focused most of my attention on methylation, environment, and inherited traits. Is there much data linking methylation and aging? Is it showing cause or coincidence?

    Thanks for this video. My friend tried it but said it had 0 effect on his running speed or ability (as a functional indicator). Like myself and a lot of females, bodybuilding benefit isn't really of interest to me. I liked the fact you touched on methylation biomarkers as not necessarily indicators of true clinical functionality.

      @@FadeToBlack888 yes resistance exercise is important, and muscle tone and strength but I am talking about those Heman body builders who are just doing it for looks - that kind of body building. In general studies have found if you keep exercising and doing strength training, people in their 80s can have strength almost as good as those in their 20s and 30s and can run really well too if they are athletic types.

    It's pretty interesting, but I'm really following Brian Kennedy. I think his trial will be more definitive. However, Brian is also an author on this paper.

Scientific analysis of the results should bear out the claims of a paper as true or false regardless. Most of the new innovations we're going to see in this field for years to come are going to come from companies.

      @@SlamminGraham I don't dismiss it outright. The results could be valid. However, we know that financial incentives could sway the results of any study.
      Anyway, even if there were no financial influence, it's just one small study.
      It's not something I would put a lot of stock in.
      If multiple independent trials have the same or similar findings in the future, then ca-akg will be more interesting. Unfortunately, the upcoming trial is not really independent of the current trial given they are each atied to Rejuvant.

      Yes I do.....................shingles. My wife was supplementing with arginine and came down with ocular shingles. The most miserable and painful affliction you can imagine. She now has to take a anti seizure drug because of the residual nerve pain. We scanned the web trying to figure out why someone with high vitamin D levels and who has been supplementing for years would get this. We found the answer on bodybuilding forums where health young men were getting shingles due to their taking arginine for it's reported growth hormone releasing effect. Beware - you don't want this. If you have to take it for any reason - offset it with Lysine.

    How do you reconcile the fact that in Sports Nutrition, AKG showed signs of improving muscle building by activating mTOR, and it’s added in some Sports drinks. On the other hand, In at least one longevity study that I’ve read, AKG seemed to block mTOR but activate AMPK pathways instead (e.g. autophagy)?

    Does simply changing the DNA methylation pattern truly reduce aging? The pattern is correlated with age, but the AKG intervention may itself change the correlation. This study is not at all convincing.

      When the article shows "duration of treatment is not correlated with the magnitude of the effect", so the benefits become saturated over time. Does that mean you take Ca-AKG on and off?

    Also there is no control group to compare the measurements against, there should be a group where they measure before and after 7 months to see what the changes are in DNA methylation. There also needs to be peer reviewed, 42 is also not a big number although statistically it's significant enough to predict normally distributed behaviors to 95% confidence levels, but this is assuming aging improvements are normally distributed through AKG supplements. This also needs to be replicated multiple times as well, double blind test...this isn't rigorous enough for scientific publication.

      The study was literally designed by "B.K.K", which is a BOARD MEMBER on the company that SELLS REJUVANT FOR A PROFIT. They, and not an independent scientific team, designed the entire study.*
      A person who has the biggest conflict of interest and whose job is to sell the most of a product, SHOULD NOT BE DESIGNING A STUDY THAT SAYS WHETHER OR NOT THE PRODUCT IS WORTH BUYING! Every data point in this study should be considered false until proven otherwise, and the University of Singapore should be ashamed to tarnish their reputation like this.
      Let's actually look at this study in this video:

      Multi-center?=❌
      Peer-reviewed?=❌
      Study literally designed by the very people that want to sell the product?=✅
      This study is clearly completely biased, if not totally fake. In the modern era of hundreds of snake oil products, and where only 39% of modern studies can be replicated by another source**, I'm sad to see "Sheekey Science Show", a channel dedicated to science, not only calling out these fraudulent individuals on any of these blatantly non-scientifically-rigorous, still-untested claims, but actually calling the results "very exciting." This video is nearly the same as her reviewing a "Cigarettes Are Good For You" study funded by Marlboro and calling the findings "encouraging." It's really disappointing that she's not making it clear how unprovable, untested, extremely biased, and most likely blatantly wrong, the results are.

    Some people on a forum tried that stuff and get their methylation age tested. Their "age" came down, but bounces back, when they stopped using it. The dirty cheap AAKG worked too. You need to take it more often though, because the calcium in CaAKG slows down the absorption.
    Side effect was highly elevated liver enzymes, which was counteracted by betaine and glutamine supplemention.

    I was listening to Peter Attia's podcast recently and was surprised to here him say where are no good biomarkers for aging. I know that dna methylation clocks aren't perfect, but I was under the impression that they were pretty good?! Also do you take a AKG supplement?

      The issue is we don't know whether these methylation clocks are just that, "clocks" or whether they actually play a causal role in the aging process.

      @@henvest3491 Ca and Arginine are just the binding molecules for AKG which transport it from gastro into the bloodstream, in the end it has the same effect. Even if CaAKG would have 10-20% better absorption it wouldnt justify that high price.

    Very excited that this study was finally done on humans, very slow progress in this direction taking into account that similar study on nematode worms was done almost 10 years ago. I am wondering if longevity effects are related to the ability of α-Ketoglutaric acid to bind ammonia therefore helping body to detoxify metabolic byproducts.

      Ammonia is a big problem for maintaining proper protein production, but Ca-AKG does a lot of things from correcting secondary hyperparathyroidism, which correct calcium metabolism dysfunction, to phosphrous metabolism dysfunction, to amino acid imbalance in aging which screws up IGF levels and over-activates integrated stress responses, and can help with better methylation patterns.

      No nad supps don’t work. They increase nad in the blood but not in the muscles where it’s needed.

    Okay, so this is odd. I read this comment about rejuvant from over a year ago on the age reversal network
    "The speaker presented a talk indicating that users got substantial drop in DNA methylation age results - I seem to recall an average of 8 years after 6 months usage"
    Either Rejuvant has been sitting on this data at least since 5/1/2020 or they just happened to repeat previous study results almost exactly. Or maybe it's totally normal to first publish results more than one year after a study has been completed?

      yeah I also got the impression from reading around that this study was pretty much already done. Maybe they were looking for some detail differences between the old and the new study. By not changing a lot of what is already known, maybe avoid risk to the company.

      It can take years to publish an article. It takes months to write a good paper. It takes months to get reviews back from a journal. Then you have to revise and the cycle repeats. The journal may want more than one round of revisions and the cycle repeats. If a journal declines to publish your paper, you have to revise and submit to a new journal and the cycle repeats. Once a journal accepts your paper it can take many months before it makes it into print. Some but not all journals now post accepted papers online while they wait for formal publication. It is not uncommon to wait for 6 months after acceptance to get formal publication. I've had several articles take more than two years between first submission and final publication.

    Where is the study showing that TruAge DNA methyl testing actually correlates with the age of an individual? Just because something is up-regulated due to DNA methylation and histone acetylation doesn't mean it accurately shows the biological age of an organism. C. Elegans could have a much higher methylation of DNA/Acetylation of histones due to the organism having such a short lifecycle, just because it happens in other models doesn't mean it applies in humans regardless of the conservation between disparate species.

      All the testing on CA-AKG and lifespan has been done on the Ca-AKG version - from flies and worms to mice. So - thats the only version we "know" that works (at least to the degree that the results have said it does). Its impossible to know if the Argenine version of AKG does the same thing - without repeating the tests. People don't want to do that - so we have to stick with the knowedge we have.

      Serum proteases(in your blood) and the acid in your stomach will cleave the arginine from the AKG, at which point the arginine will be broken down into nitric oxides which will dilate your blood vessels with the AKG being left as a standalone molecule. As such, if you can only find ARG-AKG (check amazon, thats where I got AKG from) then you will want to take double or triple the dose since ARG constitutes over 60-70%% of the molecule ARG-AKG by mass.

      Brian Kennedy is the Chief Science Officer of Rejuvant and Rejuvant funded this study. There is some inherent bias in any trial involving Brian Kennedy since both he and Rejuvant have financial interests in the material studied.

    ONE CONCERN: Is calcification of the arterial walls a possible danger with the Ca-AKG (Calcium Alpha-Ketoglutarate) supplements because there is a lot of caution with this possibly being the case with other calcium supplements?

      Percentage of Calcium in Ca-AKG is pretty low I guess. 1g of Ca-AKG could contain only 25% Ca. This means at most 250mg Calcium. RDA of Ca is 1g. So, perhaps we just need to reduce other sources of Ca, take Vit D3+K2 & Magnesium. This will further ensure Ca does not accumulate and K2 will ensure Ca doesn't deposit in artery walls while D3 helps Ca absorption and Mg ensures D3 works well.

    Thanks Eleanor. Yes the placebo effect, which can be powerful, needs to be accounted for. Would like to see how this stacks up against the diet/lifestyle study done by Kara Fitzgerald., which seem to show age reversal over a much short intervention.

      It's hard to imagine that a physical test can be affected by placebo. I mean mental and mood changes for sure but not physical. I can't grow bigger muscles just by placebo for example.

    What’s with the vitamin A and D? I thought men benefit from equal if not greater vitamin D levels and women don’t have vastly different vitamin A requirements from men.
    Couldn’t the inclusion of either also compromise the findings since they could just be treating deficiencies or whatever. Hope I’m missing something and/or this isn’t the only formulation that’s been used in high-profile Ca-AKG research (though since Rejuvant keeps funding it…)

    Is it not possible that AKG directly influences methylation? Were possible mechanisms for this proposed in the paper? After all, it seems much more plausible that a medication would directly affect the methylation sites that they happened to measure rather than literally reverse aging, which would mean having targeted and positive effects on a huge number of pathways, many more than would be required to just affect DNA methylation. Doesn't Occam's razor favor the latter hypothesis until proven otherwise?

    I'm considering starting to take MNM, TMG, and. AKG. Is there any worries that any of these will have an adverse effect on me. I had a bi-cuspid aortic valve replaced and I now take Warfarin. I can't find any information, and would like to make an informed decision before trying these.

      @@darkpatches yes, also resveratral. I can't find anything about the NMN. My doctor is a dead end. He is very close minded about these types of issues. His answer to everything is medication. Not very open minded about natural remedies. NMN is not classified as a"drug" per say, it doesn't even register on any of the drug interaction sites I have found. I'm probably gonna just move on from these if I can't find any information. I won't take that chance.

    Interesting, but all biomarkers of aging tests are not well tested, or compared or qualified - so this is not a ton of value. Did they use a bunch of different biomarker tests and just show the one that showed the best result?

      All these supplement interventions are still very early in their testing cycle and its still questionable about their effectiveness in the human body. The most proven therapeutic is Rapamycin, but its a drug, not a supplement.

      I definitely do all the free interventions (sleep, reduce stress, eat less with intermittent fasting, low glycemic diet, exercise, weight control), and then I hit on increasing NAD+ with NMN/NR, take berberine to control blood sugars, and then increase glutathione levels with GSH or high NACcysteine/glycine intake. Also taking as many polyphenols as I can, as recommended (resveratrol, fisetin, curcumin, quercetin, plus apigenin and spermidine). These are all pretty well backed by research to be safe and effective at improving mitochondrial health, but please do your research and speak to a doctor. I may throw in Ca-AKG now.

      @@eaappell That is quite risky qnd naive to think that a helathy body will be improved by supplementing those things. the research certainly doesn't suggest supplementation especially when combining them. many of those polyphenols might act over hormesis, meaning they even cause damage to your body which might amplify when combining many substances. glutathione isn't even orally bioavailable.

    I started on rejuvanant but came across novos core. What's interesting is that it has more a kg than rejuvanant but has about 10 other things in it each of which attacks a cause of aging. It seems like a much superior product. Check it out.

      At this point in time, it's probably not a superior product because we don't know how any one or combinations of supplements affects aging. So having a product with a singular ingredient is a better measurement to see it's effects then afterwards, we can add multiple ingredients to it. I hope I'm wrong and the other product is indeed superior in anti-aging though

      Yep. For analgesia for example, if you know you are being given a painkiller, it has twice the effect than if they don't tell you about it. And it's not even you making it up. If they give you naloxone previously, to block opiate receptors, the placebo doesn't work.

      Time release is usually obtained by compounding the drug with other molecules that hinder rapid absorption. In the pharmaceutical world, this is known as the "formulation" of a drug, i.e. the physical form of the active compound and the other substances that it's mixed with.

    No placebo, waste of time. The difference between the control group and the intervention group is the actual benefit which as things stand now is unknown.

    I have been taking arginine akg (aakg) for several months. Does anyone know if this will give the same results. Its so much cheaper than calcium akg

      My wife was supplementing with arginine and came down with ocular shingles. The most miserable and painful affliction you can imagine. She now has to take a anti seizure drug because of the residual nerve pain. We scanned the web trying to figure out why someone with high vitamin D levels and who has been supplementing for years would get this. We found the answer on bodybuilding forums where health young men were getting shingles due to their taking arginine for it's reported growth hormone releasing effect. Beware - you don't want this. If you have to take it for any reason - offset it with Lysine.

    I’ve been taking AAKG for about 10 months. I’m 65. Male. I’ve found I put on more muscle and less fat but the only side effect perhaps is my feet. I have pain on the outside of my right foot which I suspect is calcium deposits though I have not been to a doctor. I suspect the AAKG causes an accumulation of calcium in some way.
