Two points I gotta make: Don't be afraid to use extra water for the coffee bean extraction- you can always reduce down by boiling (caffeine is pretty hard to burn, you'd have to get it past 235C to even think about it). Also I feel like drying the DCM was largely unnecessary and even harmful (the salt water likely stole a significant portion of your yield). Since you are drying your solvent with heat, why worry about having an anhydrous solution?
You can boil the coffee in sulfuric acid (to free it from it's acid-bound form), make it slightly basic with NaOH and extract it with chloroform instead of DCM (both work fine probably). We used that method at uni.
In my personal experience alkaloids extract better from plant matter in an acidic environment and once filtered and the volume reduced freebasing the alkaloids into a suitable organic solvent that is miscible with target freebase alkaloids....sometimes butanol can do wonders Also if the water is holding the alkaloids you can always salt out the alkaloids using an appropriate salt..just saying their are better ways to do this extraction and get better yields...patience pays off tremendously
Did this at school in the 1960s but used a large catering can of Nescafe instant coffee. Got quite a lot of caffeine yeild out of it which went very well with the moonshine alcohol made from fermented and distilled sugar and yeast. Hell of a party that weekend.
Chemistry is fascinating. The entire time I was wondering whether the dichloromethane would be removed, and if so, how. In the end, he just heated it up because dichloromethane has a low boiling point. Fascinating that it's possible to do all this mixing and removing!!!
DCM is an organic solvent, think of this as if you were to dissolve a bunch of salt in water, then it's intuitively evident in order to get rid of the solvent you'd boil or evap the water and in the end you'll be left with salt

Remembering back alkali/alkaline earth metal carbonates are a large constituent to ash from wood fire. They are usually a bit less soluble than the bicarbonate form and may have higher ph though I never got to test that.
Years ago, my brother bought a 1 kg tub of pure anhydrous caffeine so that we could caffeinate just about anything we chose to drink. This experiment gives me a special appreciation for the processing that went into that.

I think part of the problem in the low yield may be that the brewing technique that you used didn't get all of the caffeine out of the coffee. I say this because it has been found that properly brewed pour over coffee (which is effectively what you were doing) has a higher density of caffeine than espresso.

Yeah, a group of my friends go really into it and were buying pure cofein to snort. One of them almost did OD (he was having muscle spasms, chest pain etc.) and had to be hospitalized. While it is almost impossible to OD on caffeinated beverages, it is extremely easy to do so on pure caffeine.
Copy paste Put in 10g of coffee into a container followed by 4g of sodium carbonate, then add in 60mL of distilled water (increase amount of ingredients for larger quantities) , and then bring the solution to boil for 15 minutes, stir or shake occasionally to prevent it from foaming over, or turn down the heat to simmer, once the 15 minutes is over you can let it cool but make sure to keep the temp as high as possible, and then strain it in a coffee filter, and then add 100mL of boiling distilled water, then pour the mixture into a separatory funnel and then add 15mL of dichloromethane, and gently shake it around, after that pour out the DCM which has the caffeine in it, and repeat the process with the remaining coffee, and then pour the DCM into a clean separatory funnel along with a saturated salt solution to pull water from the solvent, and then gently shake it again before pouring out the DCM, and add in drying agents like molecular sieves and dry for 20 minutes before placing it into a new beaker, and the boil to remove the DCM, but don't overheat it as it will burn, and then, once dry, recrystallize the remaining substance with a 95% ethanol solution, and then let to rest in the freezer before vaccum filtering and letting it dry in the vaccum for a few minutes.
Nice video! We compared caffeine content years ago at varsity on different beverages. Contrary to popular belief, a shot of espresso had the lowest caffeine content of everything we tested (lower than tea, instant coffee and perculated coffee, etc). This was mainly due to the short infusion time when extracting espresso from the machine (20 seconds). The type of roast also influenced the caffeine content - the darker the roast, the stronger it tastes, but the less caffeine it contains due to longer roasting. So if you want to stay awake, leave the espresso and opt for a weak tasting, but "powerful" pale roasted perculated coffee.
I feel like there is a fine line between the right amount of caffeine and productivity since working faster doesn't always mean that the output is as productive, but it is some what better to have coffee but not specifically multiple cups a day

You said that there is a possibility to make an emulsion. Well thats true, but the emulsion is not stable and will disperse again after a short time, overwise use a bit more DCM. You can also cook the coffee for a longer time, those 15 minutes are not enough. Also try to invest into a Rotavapor, just cooking DCM is not a good idea at all (I hope you got some air-filtering unit above it or a digestor). We use Sodiumsulfate instead of a molecular sieve as drying agent for the DCM. We didn't recrystalise, because we made titrations with it. Reason is also because its purity won't be that good (far from 99%). Cheaper to just buy the pure caffeine from your trusted merchant / pharmacy. but nice vid!

why use codeine? it is HORRIBLE.. it has a ceiling limit of 300mg, it causes itchyness and possible hives not to mention headaches.. just use hot water and lemon juice to extract morphine from poppy seeds.
Or if you are the store bought kinda guy, you could try DXM. Be careful as to what product you buy though, as some have other active ingredients which will harm or kill you if you're really lucky.

You should try this experiment again with chloroform as the solvent. I am planning out the experiment and still have to make more chloroform. Caffeine is apparently 2 times more soluble in chloroform than in DCM. The only downside would be the fumes from boiling, but you have a fume hood. I think it'd be interesting since basically everybody uses DCM for caffeine extractions. Well, I tried it with caffeine pills and it worked really well. I got a relatively low yield because I was sloppy and a lot of the caffeine was lost on the walls of the containers after the chloroform evaporated off. I didn't have enough chloroform to do proper washings. Still ended up with about 8 grams of impure caffeine. I'm going to recrystallize it from water tomorrow.

One trick I learned years ago with coffee is hold the filter firm with one hand and sharply tap the side of the thing with the other fingers, a sharp rap will make the water go thru faster.

The lighter the roast, the more caffeine content. Medium to dark roasts are roasted longer and as a result have more caffeine burned off for a more full-bodied coffee experience.
To expedite the filtration and minimize coffee loss I would recommend dampening the filter with DI water. Wet it after you begin brewing and it should be damp but not soaked.
Just found this video. I normally make my coffee with 20 g of beans to 300g of water. Plus I know that lighter roasts usually have higher caffeine content since less is burned off in the roasting process. Together, my business partner and I own a coffee company, maybe we could send a light roast coffee to you to see if it yields any more caffeine than the Folgers.
+fpm1979 I should have mentioned that! If the goal is to get caffeine, they just make it. If the goal is decaf coffee, there are several. ways. Two commons ones are super-critical CO2 and using methylene chloride directly on the coffee beans.
10 grams of coffee makes how many cups of coffee? If it's about one, your yield seems pretty great. Periodic videos extracted caffeine from 6 cups of coffee and got around 100 mg. That's about 16.6 mg per cup for them, your 30 mg looks like a much better yield. Unless they had an unusually low yield, your yield seems a lot higher.

Hi NileRed! I've recently been using a supplement called licorice root; but it has glycyrrhizic acid which poses some health problems with long term use. There is such a thing as a deglycerrhizinated licorice root. I've recently procured pure licorice root powder and I wanted to know how you would deglycerrhizinate it in the lab. Thanks!

why use codeine? it is HORRIBLE.. it has a ceiling limit of 300mg, it causes itchyness and possible hives not to mention headaches.. just use hot water and lemon juice to extract morphine from poppy seeds.
Or if you are the store bought kinda guy, you could try DXM. Be careful as to what product you buy though, as some have other active ingredients which will harm or kill you if you're really lucky.

You should try this experiment again with chloroform as the solvent. I am planning out the experiment and still have to make more chloroform. Caffeine is apparently 2 times more soluble in chloroform than in DCM. The only downside would be the fumes from boiling, but you have a fume hood. I think it'd be interesting since basically everybody uses DCM for caffeine extractions.
Well, I tried it with caffeine pills and it worked really well. I got a relatively low yield because I was sloppy and a lot of the caffeine was lost on the walls of the containers after the chloroform evaporated off. I didn't have enough chloroform to do proper washings. Still ended up with about 8 grams of impure caffeine. I'm going to recrystallize it from water tomorrow.
Lol I’d personally be terrified of running a reaction using chloroform, especially without good access to a fume hood.
Are there other solvents that could do the job that *wouldn’t* knock me out if I slipped up? 😂

1 shot of Espresso (10 grams of coffee grind -> 15 grams of liquid) contains about 30ish mg, so your yield is pretty damn amazing tbh.

One trick I learned years ago with coffee is hold the filter firm with one hand and sharply tap the side of the thing with the other fingers, a sharp rap will make the water go thru faster.

The lighter the roast, the more caffeine content.
Medium to dark roasts are roasted longer and as a result have more caffeine burned off for a more full-bodied coffee experience.
To expedite the filtration and minimize coffee loss I would recommend dampening the filter with DI water. Wet it after you begin brewing and it should be damp but not soaked.
you could try using robusta instead of Arabica coffee. because robusta contains more caffeine

Just found this video. I normally make my coffee with 20 g of beans to 300g of water. Plus I know that lighter roasts usually have higher caffeine content since less is burned off in the roasting process. Together, my business partner and I own a coffee company, maybe we could send a light roast coffee to you to see if it yields any more caffeine than the Folgers.

+fpm1979 I should have mentioned that! If the goal is to get caffeine, they just make it. If the goal is decaf coffee, there are several. ways. Two commons ones are super-critical CO2 and using methylene chloride directly on the coffee beans.
10 grams of coffee makes how many cups of coffee? If it's about one, your yield seems pretty great. Periodic videos extracted caffeine from 6 cups of coffee and got around 100 mg. That's about 16.6 mg per cup for them, your 30 mg looks like a much better yield. Unless they had an unusually low yield, your yield seems a lot higher.

That is true. I kind of did it and then realized it was the same, ha. Also, I actually dont have a coffee maker at home anymore...so that makes things more difficult.
Nile Red Ah, that is unfortunate. You kind of did what a coffee maker does anyway though, run hot or boiling water through ground coffee. You'd just have to adjust your measurement to a cup.
