Nope, I am very sure that -chan can be used for either boy or girl, and it is used to imply an intimate relation, which is why people who are not close to one can be irritated when called with -chan (and also, you are thinking about One-chan, not Nii-chan, which is a cuter version of Oni-chan, the other version for One-chan is Nee-chan)

They just noticed they had paws....then they thought only both of them has paws since they're related...then they found out that the lion has paws as well in the end....so BB asks if they're related with the lion as well....... I....don't really know Japanese soooo I'm probably wrong but I think that's what happened in the video....
