Tyson is a beast. I would say the final three bosses in Ninja Gaiden. The bosses themselves aren't really that bad, but the penalty for diyng at any point is going back to stage -1. I would also say that the Dracula fight in Castlevania 3 is pretty tough also.
I recently played through Castlevania and forgot how hard Dracula was. I think Mike Tyson took the longest to beat. I could put the code in to get to Tyson by muscle memory because I put it in so many times!

I found the duplicate mini boss in Megaman 1 to be a very challenging mini boss!
