    I reduced the solution I had to about 10mL than added 300mL of water. This caused the solution to become very light yellow but cause the curcumin to come out of solution. Lots of is stuck to the beaker I was using and the rest in the filter. I washed the walls of the beaker and the filter with alcohol to recollect the residue than reduced that. It got rid of all the water soluble crap.

    Seems after evaporation essential oil is also there in addition to curcuminoids.

    How would the end process work in regards to making a tincture? Since tinctures are supposed to be liquid, would you just take the end product in the bottom flask once it runs clear, and that would be the tincture?

    Will the curcumin not be degraded or lost its functional activity by such heat as much as 78 degrees?

      Additionally, you could use a vacuum assisted soxhlet extraction, but that requires a bit of extra gear. Namely a vacuum pump, vacuum regulator, and water chiller(or ice).

    Did you analyze the curcumin in the photometer before separating it from the solvent?

      is that a magnetic stirrer ur using?
      and what do u mean flushing air to it
      is there any other way to separate curcurmin from the crude product other than column chromatography

      1)yeah it's indeed a magnetic stirrer equipped with a hot plate, pretty useful in chemistry!
      2)The very last traces of solvent left within the final product are usually removed by terms of a high vacuum system equipped with a liquid nitrogen trap, as I don't have one, I remove the traces of solvent by blowing a current of compressed air on to the surface of the solution (ideally it should be nitrogen instead of regular air)
      3)Column chromatography is by far the best option in this case, however sometimes we can use other alternatives such as crystallization, liquid liquid extraction or precipitation..

      Well , in a professional lab , we use a lab air compressor connected to a dessicator , however i use an aquarium air pump(they are much cheaper)

    In your video,you have shown the yield as 1.1 grams for 10 grams of sample which represents 11% yield. Also from the UV spectrum,the absorbance value is found to be 0.6(approx).Using this absorbance value,i have calculated the % of curcumin which came out to be 3.5%.Why is there a variation in these values?

      Have you considered the fact that the solution measured is a diluted one?

      @@chemistry4all520 can you please elaborate on that?. Actually i have taken the standard value of 0.42 as absorbance of curcumin at 425 nm.

      Yeah that's the point, without that info is imposible to calculate how much total product is actually obtained. Unfortunately I don't have the dilution factor nor the amount of sample employed.

    The dryed extracted compound, in which what typebuffer or pH is used to maintained the stability of the extracted solvent, with out buffer it is not degrade

    You used Ethanol for this process. So I have some questions. After the Soxhlet process you have to distillate the solution again, then you get Curcumin and the Ethanol back (could be the same amount or less). So my question is; does the Ethanol after distillation contain any aroma of Tumeric?

      kirykosal, that's a very good question indeed!,experimentally the ethanol obtained is as pure as you can get (with the original azeotropic composition) but just in case some tiny impurities ended up in it as it passes through the distillation setup I added some activated charcoal so that I can recycle the ethanol. It might be possible, though, that some volatile compounds are codistilled with the solvent, but in the case of turmeric I think that most of the compounds are solids or have a very low vapor pressure so codistillation is not a huge issue

      Thank for a very clear explanation. But I would like to know too; if I want to obtain the optimal aroma (in this case) in the distillate, what should I do in this case? (I want to use the distillate not the extraction). Or it should be work with other methode? Do you have any plan to do clip on that topic? I am interrested in that topic.

      +kirykosal I'm not sure if I understand your question, if you are interested on an essential oil,there are several methods that you could use, carrying out an extraction is rather non selective and it would probably require a previous knowledge on the properties of the compound itself. A very trivial way to extract aromas is by using steam distillation (I indeed uploaded a video on limonene extraction using that method)

      Hey!, the turmeric that I used was sold as a finely grinded powder, so I cannot help you with that, but I guess that it can be easily grinded using a regular coffee grinder..

      i want to extract curcumin from javanese turmeric. i have all the equipment needed(except spectrofotometer thingy) in this experiment. but the only problem is i dont have the time. You see im a high school student and i have approximately 8 hours of school plus 45 min of after school. i can use the time given by the school for soxhlet extraction bcoz the video said it takes 8 hours to extract the substances inside turmeric. but i dont have enough time for the the distillation process (you said 2 hours). now my question is can i store the result from the soxhlet extraction in the fridge or just room temp?

      Yeah , of course there should not be any problem at all , i have stored curcumin solutions (in ethanol) at room temperature for more than a year and i did not notice any kind of degradation

    For the isolation of curcumin among three components ...Which mobile and stationary phase should be used by column chromatography??

      I've not performed the column chromatography of these 3 compounds, but I would recommend using SiO2 or Al2O3 as stationary phase. Regarding the mobile phase, I would suggest you to try different mixtures (hexane, ethyl acetate, diethyl ether..) via TLC to select the ideal one.

      Hey!, well actually the temperature of the system is given by the reflux solvent used, that is , it is determined by the standard boiling point of the solvent that you are using , since i´ve used ethanol , the temperature is around 78 ºC.
      And about the solvent , sure you can use acetone instead

      No, as I said in the video, the product is probably a mixture of 3 components.Further purification by means of column chromatography should be used to Isolate the pure product.
