  • Capsaicinoids are interesting compounds with multiple, very different uses.
    In this video I use a Soxhlet extractor to extract the capsaicinoids from Ghost Chili peppers.

    1. I basically just boiled a bunch of dried chilis.
    2. In a tiny, thin walled pot.
    3. In the kitchen.
    4. I then left the long enough for the water to boil off completely.
    Cue burnt chilis, and noxious smoke cloud in family kitchen!
    But hey, 22 years later, and now I know how it's actually done! Thanks! 😁

    The next step for complete purification should be running a silica gel column, since one of the stated impurities is beta carotine. Carotine is completely nonpolar, so starting with hexanes should flush it out. Then you can gradually increase the polarity of the solvent to grab the capsaicin, which has several polar functional groups.
    I recently did this to spinach to remove carotene and pheophytin from the chlorophyll, which was my target.

    • @abidababida7096
      @abidababida7096 10 років тому +29

      So is this next step necessary for consumption? or just to purify the capsaicin even more to get it as close to 100% pure as possible? I guess what I really like to know is if it's possible to take a totally harmless consumable product like a pepper, coffee, or even a flower like a dandelion, use an extraction process like this and come up with something that can kill u?? Obviously there are things out there that have some pretty harmful chemicals within.

    • @NileRed
      @NileRed  10 років тому +64

      Abida Babida No it is not necessary. It is merely to try and obtain pure crystalline capsaicin. However, you actually should NOT eat pure capsaicin because your body might respond to it dangerously. Assuming that you completed the extraction at home and with food grade materials, then this extract is perfectly safe to use in food.
      Using this extraction on many safe foods can be dangerous. Some foods have chemicals that are safe in small doses, but toxic in higher doses. The amount of this chemical in comparison to the volume of food keeps the dose low. By concentrating it, you can get to a level where it is unsafe. It really depends on the food that you choose and the solvent that you choose to extract it with. In general though, you should always be careful and fully research everything. Especially if you are planning to eat your product.

      Nile Red Well your answer just made me think n realize that however much this interests me, the fact that I'm not a chemist n don't know much about chemicals tells me I can wind up doing something stupid & dangerous if i don't educate myself on this more. Thanks!

    • @NileRed
      @NileRed  10 років тому +43

      Abida Babida You definitely need to research everything before you carry out a reaction, but that is how you learn. You can watch videos online and read background about various reactions. Just from this you will learn quite a bit.
      What type of chemistry are you more interested in, organic or inorganic?

    • @Matt_Deluca
      @Matt_Deluca 5 років тому +84

      I've done this before. Started with 50% fine ground espresso and 50% breakfast blend. Added it to a 500 mL soxhlet with about 300 mL distilled water in a flat bottom flask. In this instance, each extraction takes roughly 1-1.5 hours. I've found that 2 or 3 extractions yields a very delicious product with a nice "wake me up NOW" potency. 4 or more extractions, however, begins to bring a lot of undesirable flavors over. By the 5th extraction, it's almost too bitter to drink. One thing worth noting is that after the first extraction, when the water/ coffee returns to boiling it will froth up quite a bit. To combat this, I use an oversized boiling flask and strong magnetic stirring to keep the froth from going up into the soxhlet. Expect to spend several hours to make 1 cup of coffee. It will be very good coffee though as long as you don't overdo it.

    • @chrisv3593
      @chrisv3593 5 років тому +13

      That is similar to how percolating works. Ever had percolated coffee? It sucks. It's burnt bitter almost chewy coffee. In order to do this with coffee you would have to adjust pressure so it boils lower so the temperature doesn't burn the coffee or release tannins in the coffee.

    Hi there, I think the ivory solid you were left with in the 500 ml rbf was actually capsaicin whereas the dark orange syrup that you have obtained was a mixture of mainly beta carotene and "syrup-trapped" capsaicin. It might well be possible that during concentration under reduced pressure the solubility of capsaicin in ethanol has dropped and capsaicin has partly crushed out. I like your videos!

Then I cut them into ten-strips and wrapped them in tin foil and then when I’d have friends over; left them out on the table to test if they were actually my friends. Officer Meow Meow Fuzzyface
      That's not true. Well, the second half isn't true.
      All ethanol is poisonous to tissue, but it's a matter of degree. Lower concentrations need to contact the tissue for much longer to display toxic effects.
      I think the effect you're referring to is based on osmotic pressure. The less water in the ethanol, the stronger its "pickling" effect is on tissue. Basically, water flows from high concentration to low concentration. The larger the concentration gradient, the stronger the effect. Because of this, 100% ethanol "sucks" the water out of your cells, killing them very quickly. But the same can be said about 99% ethanol.
      The reason you always see 99% ethanol and never 100% ethanol is because ethanol dissolves water very easily. Even in dry climates, unless you have 0% humidity, the ethanol will promptly dissolve water from the air, and dilute itself from 100% to 99%. The near impossibility of keeping ethanol completely anhydrous is the reason you can never find it.

    I did this a long time ago with everclear. And I intentionally didn't concentrate it all the way. I dipped my whole finger in the alcohol, put it on my tongue and felt like I just ate the sun. I was also dry heaving, coughing, watery nose.... it's very powerful stuff EVEN IF dilute. If you wanna use for self defense (I was attacked by a pitbull once) just put it in a small cologne bottle. You may also be able to add some liquid butane as I think cologne bottles lock pressure unless pressed down. To keep them from diluting on the shelf. I'd just use a small thick bottle if you do this. Butane does create pressure but very low compared to other aerosols.

    • @louistournas120

    Thanks for the very informing video. I understand that extracting pure capsaicin would be troublesome given that there are other capsaicinoids present in peppers, but I was wondering if it would at least be feasible to separate the beta-carotene from the capsaicinoid mixture. For instance, wouldn't using sodium hydroxide solution (pH>11) work in an extraction procedure, since the -OH group bonded to the benzene ring in the capsacinoids would become deprotonated, thereby making capsacinoid salt that would be soluble in aqueous solution? The aqueous layer can then be isolated and acid can be added to restore the protons on the capsacinoids. I would think that the precipitated capsaicinoids can then be filtered from solution.
    If it isn't too much trouble, can you try it out and see how it goes?

    • @HeaanLasai
      @HeaanLasai 6 років тому +7

      The way I learnt is that it has to do with pain gating. The body has a mechanism for figuring out which nerve signal is correct when several nerves signal from the same area signal, a mechanism called gating. One triggered nerve will inhibit the signals from another one. By triggering the burning heat receptors, these signals are seen as more acute than pain, and the deep pain receptors are blocked. The ointment also contains menthol, which trigger cold receptors. The brain never gets the pain signal, since the pain is blocked in a peripheral gate coupling. It gets both extreme heat (capsaicin) and cold (mentol) signals at the same time, which it interprets as an ok feeling.
