Doc Oct's new robotic octopus tentacles are more than likely upgraded by absorbing the nanotech from the Iron Spider suit and that's why we see Peter in a different suit later on in the movie.

I'm so hyped for this... I loved Tobey because he was the first live action spiderman and i adored those movies as a kid, i liked andrew's spiderman because he had the wit. I like Tom's because he embodies the innocence and sweetness that spiderman has.

The best way to have a sixth member is to have the actual sixth member from the comics, or a pretender. That pretender is probably a variant of Kang, or a short time minion who serves him, either by force or voluntarily, like Wanda, like Doctor Doom, or even the Spiderman bot that looks exactly like Spiderman.
