SYNAPTIC TRANSMISSION IN THE CNS; PART 2 by Professor Fink

  • This is Part 2 of Professor Fink's Video Lecture on Synaptic Transmission in the CNS. In this Lecture, Professor Fink first describes [1] the mechanism of action for the Catecholamine neurotransmitters, [2] inactivation of catecholamines by Monoamine Oxidase (MAO), and [3] the use of Mimetic Drugs (such as ephedrine & amphetamine). Reference is made to adenyl cyclase, cyclic AMP (cAMP), Phosphodiesterase, and Phosphodiesterase Inhibitors (caffeine & theophylline). Professor Fink then describes the action & function of Inhibitory Neurotransmitters (including serotonin , 5 hydroxytryptamine, & endorphins). Reference is made to nitric acid (NO2), how foods high in certain amino acids may affect neurotransmitter levels in the Brain, the action of morphine, and the roles of Inhibition in the CNS, including sleep cycles, permitting sensory discrimination and permitting fine motor control, Attention Deficit Disorder (ADD), seizures & convulsions, and tetanus (lockjaw).

    professorfink would you say that caffeine's phosphodeisterase inhibition plays a stronger role than it's adenosine receptor antagonism in it's "energy" producing effect?

    Sir, If epinephrine is a sympathomimetic and increases heart rate, electrical conduction, contractility, and constricts blood vessels and a phosphodiasterase inhibitor is suppose to increase those effects by not allowing the breakdown of cAMP, then how would a drug such as sildenafil cause vasodilation. Phosphodiasterase Inhibitors are contraindicated when giving nitroglycerine for chest pain due to the combined vasodilatory effects.

      Firstly, epinephrine constricts most blood vessels of the body (through its action on alpha-1 receptors), but dilates other blood vessels in the body (through its action on beta-2 receptors). Epinephrine activates the enzyme adenyl cyclase, increasing cAMP (cyclic AMP) in cells. I present this information on the Video Lecture: Professor Fink's Lecture on the AUTONOMIC NERVOUS SYSTEM; PART 3:ua-cam.com/video/nPQ2UYVm4Rg/v-deo.html (at about 3l:00 elapsed time) Nitroglycerine mimics the neurotransmitter nitric oxide (NO), which activates the enzyme guanylate cyclase, which increases cGMP (cyclic GMP) within cells. Sildenifil (Viagra) specifically blocks Phosphodiesterase-6, also causing an increase in cGMP (cyclic GMP) within cells. So both nitroglycerine and sildenafil cause an increased cGMP in the vascular smooth muscle cells in the blood vessels of the penis, that leads to vasodilation. Good Question. All of this is better understood after studying Pharmacology.

    I'm suprised that more emphasis wasn't put on GABA as many emergency drugs (benzodiazepines) are given to treat seizure activity in the brain
