Just curious. Instead of modifying the trans mount why not just use a shorter rubber bushing?
It's a valid idea. I wanted to use the original parts I already had. But I don't see a problem with that idea.
How much of a difference would this make on a 727 with 4.10 rear gears? Cruising about 55mph I’m running about 2600-2800rpms and I was wanting to add a gear vendor
It's a 17% RPM reduction. So you'll be about 500 RPM below your current situation. Hope this was helpful and good luck

Do you recommended the gearvendors be used with a freshly built motor or a motor that has been broken in?
It will be fine with either. Follow standard break in proceedures for a fresh engine.
I though these were drop in and go types?
I'm not exactly sure what you mean by drop in unit. you are basically adding a backhalf to the existing transmission.
They are bolt on to some degree. The car isn’t designed around this unit, so a couple of dimples depending on your car model.
