The question is full and correct without the "anyway", but adding the "anyway" makes it also say that they already talked about it earlier too. The effect is, it's not that they just don't know it, it's that they *REALLY* don't know it. (also, there is a different sense of "anyway", the "I don't care if it's raining, I'm going anyway", but that's a different one) We could get the "anyway" like this: -I was reading the paper but I don't understand this part, whose line is it? -Oh, that's the part where they can't make the door open, I love that part! (minutes later) -... OK, that's really funny, but, um, whose line is it anyway? I don't even know! -Oh, me neither, I just thought it was hilarious!

There was a famous play in the 80's that got made into a movie titled "Whose Life Is It, Anyway?" about a sculptor who became a quadroplegic and then fought for the right to end his life. The title is a line from the play, when he's arguing with a doctor. The "anyway" has nothing to do with returning to a subject in this case. Rather, its meaning is emphatic rather than indicative - it's meant to emphasize the point of the preceding clause. The name of the TV show is a pun on the title of the original play. Thus, "anyway" just reinforces the "whose line is it" part. (as there is only the title, it's impossible for the "anyway" to be referring to something previously said.)
