    Super glad to have found this! I did a ton of work with recombinant DNA a few years ago (since then have been on a materials science/design engineering journey) and I was looking to refresh myself on the concepts I was immersed in not too long ago. This was perfect and should honestly be shown to every fresh student in a synbio lab.

    5:40 fyi You can also use the white-blue verification to see if the recombinant Plasmid is working. Here the MCS, the part where the DNA is inserted, lies within the promotor region of a LacZ gen, that will turn the colony blue, if the medium they grow on contains GalX. If the insertion was successful, LacZ cannot be produced, which means the E.Coli colony remains white. Sorting then, is simply scraping the white colony of and to proceed cloning with that. (Works only after you already pre-selected via Antibiotics)

      Exactly. In my food microbiology class we're currently learning about precision fermentation and this video just summarize the entire 40 slides.

    If a gene is inserted incorrectly in the plasmid, will RNA POLYMERASE start transcription or not? The bacterial promoter works but rna polymerase will start transcription?
