    Hi klara. Can you help me with my calibration curve I used shimadzu but I never used agilent. I want to use my hplc agilent 1100 series in my lab but I want to know how to set up my calibrations and calibration curve. I know how to set up my vials with all proper dilutions I just need to know how to run the vials and how to create the method from scratch.

    Hi Klara, you are using a 1100 system which is no longer in the market. I am going to buy a used one for my lab. It seems the autosampler is not temperature controlled🤔. Could you please explain this. How about the pump reproducibility in gradient methods? Does it generate reproducible retention times? Thank you

      Hi Mehdi, the new Agilent 1200 Infinity replaced the 1100. It has temperature controlled autosampler. It is only important if your samples are not stable at room temperature. The lack of temperature control does not effect the separation as we inject only 10ul (max) sample into a 1ml/min volume of temp controlled column compartment so it gets to the thermostat temperature very quickly. The Agilent gradient control is one of the very best on the market in terms of reproducibility and dwell volume. We run a 3.5 min gradient from 0 to 100% acetonitrile (ran this method at GSK for over half a million compounds!) and the gradient retention time variation is in the third digit of the minute! We rely on the retention time reproducibility when we use it to derive the lipophilicity of the compounds. I bought my 1100 HPLC systems second hand for my private laboratory and they are very reliable. I have an engineer who looks after them, regular pm (every 6 month) and they never let me down!

      @@klaravalko900 Thank you so much for your response, it was so informative and useful. I am working for a Biopharmaceutical Company and we have Agilent 1260 and 1290 systems, as well as a Waters Alliance system. They are both good and recognized brands in the market; however Agilent is more user friendly but to my experience Waters has more stable pumps and is more successful in applying longer gradients. I have the same idea as yours 😀, I am going to launch my own laboratory soon.
