- Investigating the rate of dehydrogenase activity in extracts of chloroplasts. Work through this AQA A-level required practical videos. Watch this step-by-step through the writing the aim, background, hypothesis, method, conclusion and evaluation. This includes examples of exam questions and model answers.
** CLARIFICATION
Ammonium hydroxide prevents the reduction of DCPIP as it accepts the electrons instead of the DCPIP. It also affects the pH and pH will affect the proteins in the electron transport chain (NOT ENZYMES- no enzymes are involved in the LDR)

you have no idea how helpful these videos are, thank you
aww thank you ☺ So glad they're helping you
@@MissEstruchBiology Hey Miss Estruch, thanks for all the useful info. Are all these videos on the Biology a-level practical's still applicable?
Thank you... great video!
these are so helpful !! The maths and practicals are my only weak spots in my science subjects so thank you !!!
ahh that's great they are helping you! I should have all the practicals up by the end of the academic year
Very clear. Thank you.
Glad you think so 😊
Hi i have a question, i did this experiment using a colorimeter so i have values for absorbance that i measured every minute but i dont know how to lay that out in a table. I know the first column would be test tube A, B etc and the next would be absorbance, but would i put the mean of all the values from each test tube? And im not sure what comes after that. I would really appreciate a reply, thank you
Thank you very much 😊
You're welcome 😊
could i use this for my research and reference?
Does this experiment work with any named factor?
Hi, do you have a set of results?!
What is the test tube c?
Thank you for this! It's very helpful but how would you suggest drawing a table for the results for the lab books?
They general rule is that that independent variable should be in the left column and the dependent (what you are measuring) in the right coloumn(s)
You will need to find a college that will allow you to do just the practicals with them. You can use videos, like this one, to then help you write it up . I think this would be your best option
Hiya, thank you for your clear explanation. It was very helpful.
I do AQA biology but the experiment I did at school was slightly different. Are these set practicals that we have to learn the exact method for or can there be variations to the experiment?
Hope that makes sense
Hi Liv,
Yes there are a range of practicals that fit each required practical title to give schools flexibility with what equipment they may have.
This is the suggested practical from the AQA practical guide, but it is fine to have completed another
Miss what would be the colour of A and B? Would A remain blue as photolysis doesn’t occur but I’m unsure about B?
Hi, thank you for this video. I was just wondering is Ammonium hydroxide the isolation solution or is that something different? Thank you
they're different things - ammonium hydroxide is the variable, the isolation solution was just used to seperate the chloroplasts from the spinach leaves. hope this helped :)
Thanks for answering Lucy.
Thats exactly it
why cant we use diff conc of ammonium hydroxide for the independent variable
You could, but from my experience when students have chosen this variable they didn't see any difference between each experiment, as any presence of ammonium hydroxide had the same impact.
Do you have a video on hill reaction?
hello 👋🏻, this is the Hill reaction, but i just didn't use that term as it isn't used in AQA.
Hi, Is there an answer for why you would blend it for a short amount of time? Thansk
So that you don't blend to the point of causing damage to the chloroplasts/chlorophyll 😊
I wanted to revise the required practical's so wanted to know if you have covered all of them?
Hello
Not yet I'm afraid. I'm only about half through. What I have done is in the practicals playlist ua-cam.com/play/PLOfYYgIrtVMi16TQgnJsrewpgH2xaEJsT.html
@@MissEstruchBiology Ok thank you, what do you think is the best way to revise for practical's?
You could try looking through the practical skills and apparatus and techniques part of the spec and make sure you can describe each technique and explain the importance of using certain pieces of equipment. After that, look through past papers for exam q practice.
@@MissEstruchBiology thank you!
Thanks for this
However for no change, its better to record ‘No OBSERVABLE CHANGE’
Is this for OCR as well ? :/
The OCR specification for the photosynthesis required practical states:
'practical investigations into factors affecting the rate of photosynthesis.
To include limiting factors in photosynthesis with reference to carbon dioxide concentration, light intensity and temperature, and the implications of
water stress (stomatal closure)'
So this practical does include light intensity, but there is probably a more specific one for OCR on your specification/practical work book.
Can I ask why the test tubes were placed in a water bath with Ice?
to slow enzyme action to prevent enzymes damaging any organelles exposed after homogenization of the cell
Miss what about the rat practical?
There aren't any rat practicals in A level Biology
@@realstarboy aqa actually
tf is a rat practical
@@Mrh.h4ckz 🤣no idea. im curious to what his school been doing.
