Lower slots might only get 2 lanes or pcie 3.0 when its a 4.0 Chip or just sata. But if youre not doing straight data transfers but normal everyday usage like most people do you wont notice a big difference between even 4 lane pcie 4.0 and sata if they both have decent controllers since youre not limited by data tranfer rates but how fast the controller gets the data from the nand. Typical ssd operating speeds when gaming or browsing are in the hundreds of MBs.

USB 3.0 Gen1 5Gbit=625MB/s (theoretical limit, usually far less).
      M.2 SATA3=500MB/s
M.2 NVME= depends on generation, but in theory yes there will be a cap to the maximums (random read/writes will be under the caps as SSD's are often marketed well above what they'll ever see in normal usage).

@@tomstech4390 m.2 sata 3 speed is 600MB/s according to the spec. Normal sata drives dont go over 520-540mb/s due to loses with sata cable etc.

M.2 raid controllers. You can create multiple arrays for separate storage or spanned or raid 0 if your a speed nut.

Asus makes a nice PCIe adapter for 4 NVME drives into a single full size PCIe slot. Comes in Gen 3, 4 and 5 too.
Got one of them in my homeserver where I'm running the 4 drives in ZFS, works wonderfully.
