From what I know about PARPi, they actually inhibit the 2nd option in case of HRD (homologous repair deficiency), as to making the cell die of too many mutations. In case of HRD, the cell would normally rely on PARP repair, which repairs the DNA but with flaws. So as to avoid the flaws in the repair mechanism, PARPi is given as treatment.
