Hi, to find your unknown concentration you need to dilute your protein standard with your lysis buffer/diluent and measure their absorbance values and draw your graph based on your absorbance vs your known protein concentration, obtain the line equation from that and then you can measure your unknown protein absorbance and use that value to put into your line equation. These values in the video are just for illustration purposes.
if you use different decimal places/or if you round the values for the manual calculation below, it can give slight differences. If you want to be more exact you can use the 1st method shown. Thanks
