    I think the representation of -PO4 and -OH group at either end (any one) is wrong (0:57 min). You need to swap the functional groups at one of the two ends to maintain the 5' to 3' orientation correct. But you corrected it afterwards at 1:10 min. The video is very useful by the way.     Something to comment, polynucleotide kinase use dNTP labelled in the gamma phosphate, not alpha. This last one is used by polymerase.

    Sir how oH is left behind when we remove phosphate ?
    Nucleotide have 3 phosphate the phasphatase removes all the three phosphates or only one phosphate ?

    1.40 , Polynucleotide kinase only acts on 5' end , then how it add phosphate in 3' end of PCR products (As you mentioned)🤔🤔🤔🤔🤔🤔🙇🏼‍♂️🙇🏼‍♂️🙇🏼‍♂️🙇🏼‍♂️

    I think the PCR product does contain 5' phosphate group and does not require to be phosphorylated after PCR reaction by polynucleotide kinase. Please correct me if I am wrong.

      No you are correct in your point.... the trick prople use in lab is to put phosphate group also in the 3’ end using pnk so both 3&5’ are phosphate and the vector you have to use you use alkaline phosphatase to cleave 5 prime phosphate and as a result the efficiency increase
