Thats an excellent question. TF binds to specific UAS sequence and not any UAS, hence highly specific in expression of reporter gene. Also, negative controls are always used.
If we place both of the proteins in one plasmid, we also need to place the AD and DBD in the same plasmid. That means the domains (AD and DBD) are already in close proximity, leading to the recruitment of transcription machinery and ultimately results in the expression of the reporter genes. We can,t study the Protein-Protein interaction by placing both the proteins in one plasmid.

you could counter-select using the Reverse Yeast 2 Hybrid Screen. Look it up
