    In the lab, we can use molecular tools to cut and paste pieces of DNA from different organisms, allowing us to study gene function or even transform bacteria into living factories! These powerful molecular cloning techniques are used daily in research labs around the world, but simple enough to be performed in your classroom laboratory.     In this workshop, we discuss the ways that we can use molecular tools to combine genetic material from different origins to build recombinant DNA molecules. We can then use that recombinant DNA to program bacteria to make antibiotics.
    In the pharmaceutical industry, many drugs are produced in living cells in a process called fermentation, which can be used to manufacture products as diverse as biofuels, biopharmaceuticals and fine chemicals. The process of growing and purifying industrial products in cells is known as bioprocessing. Upstream bioprocessing uses genetic engineering and recombinant DNA to produce large amounts of important proteins, converting cells into living factories. Once fermentation is completed, the desired molecules must be harvested and purified from the culture. This practice is known as downstream bioprocessing. The product (in this example, a medication) is purified from the cells using centrifugation, filtration, and chromatography. When purified, the product can finally be utilized for commercial and/or industrial purposes. Insulin, which is a hormone used to control diabetes, was the first medication for human use that was produced by genetic engineering. Biologic medicines, such as antibiotics, interferon and blood clotting factor VIII, have helped save millions of lives and improved the quality of life for millions more.
    In this video, we discuss Edvotek Kit 301, Construction & Cloning of Recombinant DNA. In this experiment, students build a recombinant DNA molecule from a plasmid and an antibiotic resistance gene. It’s a long experiment, so even under the best of circumstances, you’re doing the experiment over multiple days or weeks. But, with current hybrid schedules, you may only have students once per week, or you may need to run the experiment with two sections, one on Monday and one on Thursday. So, how can time the experiment right so that every section has the best success? In this live stream, I point out the best stopping points, which reagents can be prepared in advance, and non-negotiable steps.

    The best video I've ever watched!!! Thank you!! Just one question: Where could get a step-by-step experimental protocol to produce insulin?

      Thank you so much, we are glad you enjoyed the content! We do not have a step by step protocol for producing insulin. I'd start searching on PubMed (pubmed.ncbi.nlm.nih.gov/). Remember, the gene produces a full length protein that is processed 2x before it becomes biologically active!
