  • Practical investigation of the effect of alcohol on cell membranes. Beetroot sections are placed in increasing concentrations of ethanol in water. After a few minutes, the presence of red/purple dye in the liquid would indicate that beetroot cell surface membranes had been disrupted by the ethanol. Ethanol is a polar organic solvent and can interact with phospholipids, disrupting their organisation as a bilipid layer and thus causing cell contents, including the dye, to be released into the liquid surrounding the beetroot sections. The extent of this can be quantified using colorimetry. Take care to use filters of colours/wavelengths on the opposite side of the colour wheel.     Hey quick question here! How do you Transfer the unit Abs into Au?
    Because I saw some other similar Experiments and they had huge number for the absorbance of light measured in Au?

    Great video; really helpful. Thank you. I tried doing this experiment after you and I noticed that I picked concentrations of 10% to 50% of ethanol, and as I finished writing the report, my teacher asked me why I picked these concentrations specifically, and I'm not sure how to answer that. Would you mind telling me?

      good question. but i'm afraid the answer will be disappointing! that's what came with the method I used with my students. usually, these methods are tested to show a range of their effects, from hardly any effect to a very obvious one. large ranges are usually suggested so that we can be more confident of trends in the results.

      Independent: concentrations of alcohol
      Dependent: Light absorption (using the colorimeter)
      Control: surface area of the beetroot and room temperature

    +Dr Bhavsar is the any scientific literature that explains why higher ethanol concentrations result in less damage to the cell membrane

      +Dr Bhavsar our class replicated this experiment and found that concentrations above 60, had lesser leakages of betacyanin into the surrounding solution, the teacher said this was due to the different lengths of phospholipids in the membrane and after a certain point the swapped hydroxyl groups will create a water barrier and preventing further ethanol concentration. The graph of these results is apparently meant to be an inverted v.
