The Stropharia spores germinated and were clean - the others not so much. 30% success spores to LC not that bad - the rest of the innoculations have produced healthy mycelium so far! Cannot wait to get fruits!
That's pretty impressive considering how dirty even clean spore prints are, and if you ever need a new liquid culture recipe to try I reccomend 500ml of water and 1/16th of a teaspoon of pilsen light malt extract 3.3lbs liquid. It only costs about 10 bucks and it is the best recipe I have used to date for lc, my lc is always thick and the whole jar becomes a cloud even with some mycelium growing on the surface if you don't break it up because of all the vigor. You know you did the recipe right if once the lc comes out of the pc it has no to low color change since the amount of nutrients in pilsen light is so high
Not only were you talking with the needles right in front of your face, you even took the syringes out of the sterile field several times to hold them up to the camera. You need to work on your sterile technique. There is water all over your flow hood. You understand that spilling shit all over the place isn't sterile technique, right? At the beginning of the video you waved your ungloved hand over your sterile field. WTF? Flow hoods aren't magic. You're fucking your shit up before you even start working!

I will put it on the list - my brief understanding is that it causes mutations and can therefore make new variants. Not very precise but maybe microwaving mycelium for 10 seconds here and there could also do this 🤷♂️
Good video, but 25 ml +10ml +10ml gives roughly a dilution of a factor of 2. If you want a factor of 10, put 36ml of water and 4ml of spore suspension (not solution!)
Merry Christmas! I like to use spent plates whether plastic or glass, usually I take the cracked ones when I open a sleeve and set them aside until I find the other half in another pack - if that makes sense 😄
I’m trying to understand how serial dilutions help isolate the spores away from any contamination. Is the effectiveness based on the assumption that the spores overwhelmingly outnumber contaminants on the spore print dish that you’re starting with?
it doesn’t have to outnumber contaminants, just separating all the cells until they are diluted enough will give you a chance that there would eventually be 1-2 colonies per plate - if you do enough plates then you should get some clean isolates. that’s worse case scenario

Did you do that video on snake venom. I think it's way beyond my ability, but, it's such a strange application of venom, and learning how it works via visual cues on a video medium sounds really cool. 😎
I heard once that rattle snake venom produced in a hyperbaric chamber is used as a medication. I don't know for what though. Before the Flood of Noah there use to he a Seventh layer to the atmosphere. Creating about 2.5 times the atmospheric pressure. This creates more oxygen in the air allowing for bigger animals and plants. Evidently changing the toxic nature of snake venom also. Mushrooms??

Have you got a video or any articles on how you do a sterile spore print lifting out above, using Vaseline and a jar? I've never heard of that process

Does the Polysorbate 20 come sterile, or does it need to be sterilized before adding to the spore solution?
lab grade should be sterile cosmetic grade is close but if you are careful at keeping the stock clean it should be fine

It acts as a mutagen to cause mutations. I am not sure about the exact procedures but Ill do a video on it soon
