    I want to point out that while yes, the crystal violet does eventually wash off from the Gram negative bacteria, it's the iodine that encapsulates the Gram positive first, so they don't get decolorized by the alcohol. That's when the last stain (the Safranin) would stain the Gram negatives. I understand this is an educational video, and I know some of it may have been a little rushed for the sake of getting the point across, but I believe that understanding the process of staining and how it actually works is much more beneficial to viewers than a simple, "it stays purple because that's just what Gram positives do."

    This is like 2 hours of lecture and yelling from my lecturer..do you understand class..! picture they say is worth a thousand words..thanks for d video..did a whole lot of good in my understanding..but u didn't tell the arrangements of the bacteria..single(one) doubles(two's) in chains(long or short)..in clusters(ie bunches)..anyway thanks a bunch..!

    what part of the bacteria structure holds the stain?

      pretty sure its the peptidoglycan. which is why the negative doesn't hold it well, bc its peptidoglycan is so thin

    Great video. Te only grips I had was the "wash it off" part. As we know. You stain the bacteria with ultra violet stain, keep the stain on for 30 seconds, rinse with distilled H2O, gram stain for 60 seconds, rinse with H2O, tilt glass slide at 45 degree angle, then apply ethanol to the stain slowly(one drop at a time) then rinse with H2O, Safran stain for 60-120 seconds, rinse with H2O, then blot dry, then observe over microscope.
    The application of ethanol is very important in my opinion.
    But I digress because you can't think of all the details, such as I can't either.

    Spirilla is not the plural of spirochete. Spirilla is the plural of spirillum. Spirilla and spirochetes are 2 different shapes. Spirilla are a loose coil while spirochetes are a tight coil.

      That is an error. Spirochete and spirilla do not mean the same thing. If you haven't yet, google differences between spirochete and spirilla and also check out the wikipedia page Spiral bacteria.

    This lecture is not 100% accurate sadly. A little vague when it comes to the staining techniques, to say the least. Good color differentiation in the diagram along with the black background was very enjoyable however.

    what properties does the wall have that makes the dye stay on?? does a thicker petidoglycan wall absorb the crystal violet more or something???

      Tropical StormXoX yes. The thicker peptidoglycan layer holds the dye from the crystal violet. That's what gives it the blue stain

      The thicker the peptidoglycan layer the more dye it will absorb therefore making it gram-positive. The thinner the peptidoglycan layer the less It will absorb therefore making a gram-negative

    Trick to remember that gram negative has a smaller peptidoglycan layer and a lipopolysaccharide layer:
    For athletic performance, it's negative eat more fat and carbs (lipopolysaccharide) instead of getting enough protein (smaller peptidoglycan layer)
