Hi sir thnkx for the hard work you are putting in your videos ... i have a question while we were deriving the equation of michaelis we said that when the time is nearly 0 the (Et)=(E)+(ES) but this time we said that when (S) is very low in comparison with Km we put (Et)=(E) ...isnt the same thing i mean when t is nearly 0 the concentration of the substrat is very low so how does is came that we end up with 2 differents equations ?? ... thnkx

Remli Yusupov The Km value is a property of the enzyme under some specific conditions. So by stating that [S] = Km, you're basically stating that there is a certain amount of concentration in the environment at which the enzyme is working at half its maximum efficiency.
as for a particular reaction , the amount of substrate molecule is much more higher than the amout of enzyme molecule. so therefore, you cant state that. (s)=(E)/2.

When you wrote that Vo=kcat[ES] are you assuming that the substrate is at saturating conditions? Also, great video!
