We have a "car free" day here in Paris, France, but it's mostly a political photo op on a few main boulevards, the rest of the city actually gets MORE dangerous for cyclists, because the people who still get in their cars (you just have to claim that you have no choice but to be using your car that day in case of a police check) drive even faster than usual due to the reduced traffic.
Yes, in particular FDR highway in Manhattan is closed, and BQE freeway and Gowanus Expressway in Brooklyn, and the Verrazzano Bridge to Stan Island are all closed to cars for you during this cycling event.
Consider that there are only two events in which you can experience NYC roads without cars: the 5 Boro ride and the NYC Marathon. It's also likely the only time you'll feel safe crossing the Verrezzano Bridge.

I did this last year. They literally closed off 40 miles of the city for bikes only. It was wild
