The box with Sam is a box of Seasoning salt from the Shire for Roast Chicken in the Two Towers Sam drops it and Frodo catches it and almost falls off the cliff only to find out they have made it to the bottom. Frodo says its a little bit of home.

In the books I believe the box contains soil intended for regrowing the Shire especially the Party tree. The film leaves this out
