You handled the 4g of nicotine way too casually. It's enough to kill dozens of people. Always double bag your gloves, and for the love of god get good nitrile gloves if you wanna do stuff like this. The low yield might come from the solution not actually penetrating through the tobacco properly, usually this extraction is carried out with mechanical stirring. In our procedure (from 1962) they add oxalic acid to the 1.5L distillate and concentrate, take up with KOH and extract. This always yielded very clean Nicotine in our student labs.
Nicotine toxicity was far overestimated for many years. Apparently it is far more toxic to lab animals than humans. Once vaping became a thing we had real world data on overdose fatality rates and they revised the LS50 up by an order of magnitude, as people were commonly surviving quite large doses. It is still of course toxic in large doses and dangerous to leave around children, but it is not as bad as people thought.
I've spilled 10% nicotine on my arms high enough to get it inside my gloves on my arm also. I immediately washed up and cleaned with alcohol afterwards just in case. I was fine and had probably a few grams of nicotine in propylene glycol on my skin. Not even a nicotine rush happened I actually still craved nicotine at that point lol It's dangerous but I think it's been exaggerated over time.
@@GigsTaggart The real LD50 value is likely still only somewhat higher, than for cyanide salts. The high values are from cases where people have drank nicotine, vomited and then hospitalized. IE the real absorbed dose is much lower and the survival required hospital care. Not quite the same as measuring LD50 values with rats.
In plants, alkaloids are secondary metabolites. They are synthesised by the plants via enzymes. The biosynthetic pathways are quite complex and multi-step.
Cotinine, the oxidation product of nicotine with H2O2, is much less toxic than nicotine, whilst still retaining similar pharmacological properties. It's a nootropic, and at one point cotinine fumarate was considered to be marketed as antidepressant by the pharma industry. I wouldn't be comfortable to handle pure nicotine, however apparently you can spray tobacco leaves with 1-3% H2O2, and the bulk (~80% of nicotine will be converted to cotinine). Could be to interest to those who grow their own tobacco.
a proper steam distillation isnt too difficult to set up, all you need is a flask to boil water and some plastic tubing, a thermometer adapter and a glass tube, stick the glass tube in the thermometer adapter to the bottom of the distilling flask and connect the plastic tube to the flask to boil water, or alternatively use a chromatography bulb packed with the material

I literally put all the information required into the video. The reason you use sodium chloride in water is to remove some additional water from the organic layer the exact amount does not matter.
