    please help me here
    at 4:00pm a flask of sterile broth is inoculated with 10000 bacterial cells
    at 9:00 pm the log phase culture had a population of approximately 41million cells .CAlCULATE THE NUMBER OF GENERATIONS AND THE GENERATION TIME

      Number of generations - 12
      Generation time - 29 min.

      You are correct. Generation time is 25 min. If you multiply 12 generations with 25 min you will get 300 min which is the given time period.
      For me the calculated generation time was around 29 min and number of generations was between 12 and 13 using the formula. I have rounded off the value to 12.

    If one starts with 10000 cells in a culture that has a generation time of 2 hrs ,how many cells will be in the culture after 24 hrs

    @7:45 it should be 10^7 for Nt. Check your math . G should then equal 12 minutes

    Can someone. Explain these numbers to me? Okay, let us look at this number "1,60,000" Is this supposed to be 1,600,000. This number does not make any sense. These 6,40,00 is 6, 400,000 and 12, 80, 000 is 12, 800,000.

      Multiply each product with 2, starting from 10,000, then you will get these numbers.

    The time required for bacteria to duplicate was recorded before and after mutation therapy. Observations were carried out on 15 different strains of bacteria. Apply t-test and find out whether the mutation therapy have effect on generation time.
    Before treatment generation time 19, 22, 17, 19, 15, 25, 24, 22, 28, 23, 19, 25, 24, 23, 18
    After treatment generation time 18, 14,23, 25, 17, 16, 15, 21, 15, 12, 22, 18, 19, 24, 20 ​

    Can this be used when looking into optical density as I have an assignment where we had to grow our own bacterial culture and look at the optical density over 90 minutes at 550nm? We need to calculate lag phase and generational time. I've run through some calculations and getting a rough estimated time of e-coli generational time at 26.38 minutes? (Yes I know you can't work without data but still, does this sound about right?)

    I don't know how grateful can I be to you. What do you do when you just have OD values from measurement in microtiter plates? The OD of the bacterias I am using are normally 0,286 after 10x diluted. I diluted 20x more (2 x10) Then I inoculated the plate with 100 ul of the stock in 100 ul media. The stock continues to be diluted until 10^-7. And I get a CFU/ml normally 10^9. But I don't really have an initial number of bacteria. Just the first OD which was 10x diluted.

      Produce a standard curve CFU versus OD. That may help to estimate the initial population.

      @MicrobiologyMantra thank you! I don't know if I understand well what to do. My initial values would be the number I have inoculated? For example, if my CFU/ml is 1x10^9. Could I just divide this number by 10:10:10:2? What would be the serial dilution 3x10 and 2 by the inoculum? Then I would have more or less 1×10^5?

    I have a correction:
    The bacteria did not double from 10,000 to 1,000,000. Double of 10,000 is 10,000,000.
    So it takes 18 minutes for 10,000 cells to become 1,000,000 cells.
    People might get misinformed. :)

      You are mistaken.
      I did not say that. I have taken population at two different times from the growth curve, and calculated the generation time from that.
      10^4 and 10^6 are just two different populations at t1 and t2.
      By the way double of 10,000 is 20,000.
      Hope you got me now.

      I think the time is always more than a minute, so, it is better to use in minutes.

      124 min was approximate time. Apne ko 120 min pe 10 lakh aana tha. Is liye 124 min pe 12 lakh aaya.

      This is just another form of previous equation. Just to separate the log equation. There is no change in the equation.

      Using standard plate count method after performing serial dilutions, you can get the population of cells.
      There is another method of taking optical density by using spectrophotometer. But this method has some limitations.
